目的 探讨肿瘤相关中性粒细胞(TANs)对乳腺癌4T1细胞的影响及干姜有效成分6-姜烯酚对TANs的干预作用.方法 分离及鉴定小鼠中性粒细胞,建立中性粒细胞与肿瘤细胞体外共培养体系,运用MTT法、细胞粘附、细胞划痕及tran-swell实验检测TANs对4T1细胞增殖、粘附、迁移和侵袭能力的影响及6-姜烯酚干预作用;激光共聚焦显微镜观察佛波酯(PMA)及肿瘤细胞刺激中性粒细胞后NETs释放标志物MPO和Cit-H3表达水平及6-姜烯酚干预后肿瘤细胞对中性粒细胞NETs释放影响;ELISA实验检测6-姜烯酚处理后TANs分泌炎症因子表达水平;Transwell小室法检测6-姜烯酚对PMA诱导中性粒细胞释放NETs捕获肿瘤细胞能力的影响.结果 成功分离纯化小鼠中性粒细胞;TANs能够促进4T1细胞增殖、粘附、迁移和侵袭能力,6-姜烯酚可逆转TANs的促进作用;ELISA结果显示6-姜烯酚能够抑制TANs分泌炎症因子TNF-α,IL-6和IL-1β等水平;激光共聚焦显微镜观察发现中性粒细胞经PMA或肿瘤细胞刺激后MPO和H3Cit表达增加,6-姜烯酚能够抑制肿瘤细胞诱导的中性粒细胞过度释放NETs,且可降低PMA诱导中性粒细胞释放的NETs对肿瘤细胞捕获能力.结论 TANs能够促进4T1细胞增殖、粘附、侵袭和迁移能力,6-姜烯酚通过干预中性粒细胞募集及NETs过度释放逆转其促进作用,改善肿瘤炎性微环境,抑制乳腺癌细胞侵袭及迁移.

Objective To investigate the effect of tumor-associated neutrophils(TANs)in breast cancer 4T1 cells,and the intervention role of the dried ginger active ingredient 6-shogaol on TANs.Methods Mouse neutrophils were isolated and identified,an in vitro co-culture system of neutrophils and tumor cells was established.Effects of TANs and 6-shogaol-treated TANs on 4T1 cell viability,adhe-sion,migration,and invasion were detected via MTT,adhesion,wound healing,and transwell assays.MPO and Citrullinated Histone H3(Cit-H3)was detected to assess NETs release in neutrophils stimulated with PMA or tumor cells,and the effect of tumor cells treated with 6-shogaol on the release of NETs was observed by laser confocal microscopy.ELISA was used to detect TNF-α,IL-6,and IL-1β levels in 6-shogaol-treated TANs.Effect of 6-shogaol on NETs trapping tumor cells was detected by Transwell chamber assay.Results Mouse neutrophils were successfully isolated and purified.TANs promoted 4T1 cell viability,adhesion,migration,and inva-sion,while 6-shogaol reversed these promoting effects by modulating TANs.Results of ELISA assay showed that 6-shogaol reduced the secretion of TNF-α,IL-6 and IL-1β in TANs.The expression of MPO and Cit-H3 in neutrophils was stimulated by PMA or tumor cells,and 6-shogaol inhibited tumor cell-induced excessive NETs release in neutrophils.Transwell assay showed 6-shogaol reduced tumor cell trapping by PMA-induced neutrophil NETs.Conclusion TANs promote the abilities of proliferation,adhesion,migration and invasion of 4T1 cells;6-shogaol reverses promoting effects via intervening in neutrophil recruitment and excessive release of NETs,ameliorates the tumor inflammatory microenvironment and inhibits breast cancer cell invasion and migration.
