基于LC-MS结合网络药理学、分子对接及体内验证探究"桂南"翅荚决明对鸡抗炎作用机制OA
Elucidating anti-inflammatory mechanism of Senna alata'Guinan'in chickens based on LC-MS combined with network pharmacology,molecular docking,and in vivo validation
试验旨在探究豆科牧草"桂南"翅荚决明对鸡炎症反应的调控机制.采用液相色谱-质谱联用(LC-MS)技术鉴定其主要活性成分,结合网络药理学预测潜在作用靶点与信号通路,通过分子对接及体内试验进行验证.结果显示,LC-MS分析鉴定出高良姜素、6-羟基木犀草素、桑色素、漆黄素和山柰酚等9种主要活性成分,通过数据库筛选预测得到179个潜在作用靶点;将其与2 466个鸡炎症相关靶点取交集,获得101个共同靶点.通过蛋白质-蛋白质相互作用(PPI)网络,进一步筛选出蛋白激酶B丝氨酸/苏氨酸激酶1(AKT1)、表皮生长因子受体(EGFR)和B细胞淋巴瘤/白血病-2基因(BCL2)等19个核心靶点.基因本体论(GO)功能富集分析表明,这些靶点主要参与蛋白质磷酸化、凋亡过程等生物过程;京都基因与基因组百科全书(KEGG)通路富集分析显示,其显著富集于Toll样受体信号通路、细胞凋亡等与抗炎相关的信号通路.分子对接结果显示,上述活性成分与核心靶点之间具有良好的结合活性.体内试验表明,饲喂含有20%"桂南"翅荚决明的饲粮能够使鸡血清中白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)含量降低.研究表明,试验明确了"桂南"翅荚决明调控鸡炎症的核心活性成分、靶点及机制,为其作为鸡养殖中抗炎饲草资源的开发应用提供参考.
This study aimed to investigate the regulatory mechanism of the leguminous forage Senna alata'Guinan'on the inflammatory responses in chickens.The main active ingredients were identified using liquid chromatography-mass spectrometry(LC-MS)technology,and potential targets and signaling pathways were predicted using network pharmacology.Molecular docking and in vivo experiment were used for validation.The results showed that LC-MS analysis identified nine major active components,including galangin,6-hydroxyluteolin,morin,fisetin,and kaempferol.Database screening predicted 179 potential targets,which were intersected with 2 466 chicken inflammation-related targets,yielding 101 common targets.Protein-protein interaction(PPI)network further screened 19 core targets,including protein kinase B serine/threonine kinase 1(AKT1),epidermal growth factor receptor(EGFR),and B-cell lymphoma/leukemia-2 gene(BCL2).Gene Ontology(GO)functional enrichment analysis indicated that these targets were primarily involved in biological processes such as protein phosphorylation and apoptosis.Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis revealed significant enrichment in anti-inflammatory-related signaling pathways,including the Toll-like receptor signaling pathway and apoptosis.Molecular docking results demonstrated strong binding affinity between the aforementioned active components and the core targets.In vivo experiment indicated that feeding a diet containing 20%Senna alata'Guinan'reduced serum levels of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),and interleukin-1β(IL-1β)in chickens.The study shows that clarifing the core active components,targets,and mechanisms by which Senna alata'Guinan'regulates inflammation in chickens,providing a reference for its development and application as an anti-inflammatory forage resource in chicken farming.
