试验旨在探究不同养殖模式对仿刺参肠道酶活性、组织结构及菌群组成的影响.选取360只外观完好、体色正常、规格一致的健康仿刺参,随机分为2组,每组3个重复,每个重复60只.吊笼组采用吊笼养殖并投喂海带和鱼糜,网箱组采用网箱养殖并投喂配合饲料.预试期7 d,正式试验期60 d.结果显示,网箱组酸性磷酸酶(ACP)、过氧化氢酶(CAT)、碱性磷酸酶(AKP)和超氧化物歧化酶(SOD)活性显著高于吊笼组(P<0.05).吊笼组淀粉酶(AMS)和脂肪酶(LPS)活性显著高于网箱组(P<0.05).吊笼组肠道绒毛宽度(VW)、绒毛高度(VH)及肌层厚度(MT)显著高于网箱组(P<0.05).基于16S rRNA的Miseq高通量测序分析显示,吊笼组肠道菌群涵盖16个门、112个属和89个种,网箱组肠道中涵盖18个门、108个属和65个种.两种养殖模式下肠道菌群结构存在明显差异,吊笼组的优势菌群为弧菌属、发光杆菌属、Aliivibrio属、Formosa属、假交替单胞菌属,网箱组的优势菌群为弧菌属、发光杆菌属、希瓦氏菌属、Aliivibrio属和假交替单胞菌属.研究表明,网箱养殖的仿刺参具有更高的抗氧化能力,且肠道菌群丰富度和多样性高于吊笼养殖,养殖效果更佳.

This experiment aimed to investigate the effects of different aquaculture modes on the intestinal enzyme activities,tissue structure,and microbiota composition of Apostichopus japonicus.A total of 360 healthy Apostichopus japonicus with intact appearance,normal body color,and consistent size were randomly divided into two groups,with three replicates per group and 60 Apostichopus japonicus per replicate.The hanging cage group was cultured in hanging cages and fed with kelp and fish surimi,while the net cage group was cultured in net cages and fed with formulated feed.The pre-test period was seven days,and the formal test period was 60 days.The results showed that the activities of acid phosphatase(ACP),catalase(CAT),alkaline phosphatase(AKP),and superoxide dismutase(SOD)in the net cage group were significantly higher than those in the hanging cage group(P<0.05).The activities of amylase(AMS)and lipase(LPS)in the hanging cage group were significantly higher than those in the net cage group(P<0.05).The intestinal villus width(VW),villus height(VH),and muscularis thickness(MT)in the hanging cage group were significantly higher than those in the net cage group(P<0.05).Miseq high-throughput sequencing analysis based on 16S rRNA revealed that the intestinal microbiota of the hanging cage group encompassed 16 phylum,112 genus,and 89 species,while that of the net cage group encompassed 18 phylum,108 genus,and 65 species.There were significant differences in microbial community structure between the two aquaculture modes,the dominant bacterial genera in the hanging cage group were Vibrio,Photobacterium,Aliivibrio,Formosa,and Pseudoalteromonas.The dominant bacterial genera in the net cage group were Vibrio,Photobacterium,Shewanella,Aliivibrio,and Pseudoalteromonas.The study shows that Apostichopus japonicus cultured in net cages exhibited higher antioxidant capacity,along with significantly higher richness and diversity of intestinal microbiota compared to those cultured in hanging cages,indicating a better culture effect.
