八珍散总皂苷生物提取的工艺优化及抗氧化活性研究OA

试验旨在采用植物乳杆菌和动物双歧杆菌对八珍散进行固态发酵,以提高其活性成分总皂苷的提取效率和抗氧化活性.通过单因素试验和正交试验系统地探讨了发酵时间、发酵温度、水分含量和接种量对总皂苷提取率的影响,并通过测定1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除率、2,2'-联氮双(3-乙基苯并噻唑啉-6-磺酸)二铵盐(ABTS+)自由基清除率、还原能力评价发酵前后总皂苷的抗氧化活性.结果显示:单因素试验中,当发酵时间为48 h、发酵温度为34℃、水分含量为50%、接种量为1×108 CFU/g时,皂苷提取量均显著提高(P<0.05),提取量最大;正交试验结果显示,最佳发酵参数组合为时间48 h、温度31℃、水分50%、接种量1×109 CFU/g,此条件下八珍散总皂苷提取量比未发酵时的总皂苷提取量显著提高(P<0.05),影响总皂苷提取量的主次因素依次为水分>接种量>温度>时间.且发酵八珍散总皂苷的DPPH自由基清除率、ABTS+自由基清除率、还原能力均优于发酵前,有更好的抗氧化活性.结果表明,固态发酵八珍散能够有效提高总皂苷提取量及其抗氧化活性.

This experiment aimed to improve the extraction efficiency and antioxidant activity of total sapo-nins from Bazhen San using solid-state fermentation with Lactobacillus plantarum and Bifidobacterium ani-malis.The effects of fermentation time,temperature,moisture content,and inoculum size on the extraction yield of total saponins were systematically studied through single-factor tests and an orthogonal array design.Antioxidant activity was evaluated by mea-suring 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging rate,2,2'-azino-bis(3-ethyl-benzothiazoline-6-sulfonic acid)(ABTS+)radical scavenging rate,and reducing power.Single-factor test results indicated that the maximum saponin extraction was achieved under the follow-ing conditions:a time of 48 h,a temperature of 34℃,a moisture content of 50%,and an inoculum size of 1×108 CFU/g.The increase was significant(P<0.05).Orthogonal test results showed the optimal fermenta-tion parameters were:a time of 48 h,a temperature of 31℃,a moisture content of 50%,and an inoculum size of 1×109 CFU/g.Under these optimal conditions,the total saponin extraction from the fermented prod-uct was significantly increased compared to the unfermented material(P<0.05).The primary and secondary factors affecting saponin yield during fermentation are,in descending order of importance:moisture con-tent>inoculation amount>fermentation temperature>fermentation time.Furthermore,the fermented prod-uct demonstrated superior antioxidant activity,showing higher DPPH radical scavenging rate,ABTS⁺ radical scavenging rate,and reducing power.The results demonstrate that solid-state fermentation can effectively enhance the total saponin content and antioxidant activity of Bazhen San.
