基于代谢组学探讨电针"内关"和"膻中"穴对慢性心力衰竭大鼠的作用机制OA
Exploring the mechanism differences of electroacupuncture at"Neiguan"and"Danzhong"in intervening chronic heart failure rats based on metabolomics
目的 观察电针"内关"和"膻中"对慢性心力衰竭(chronic heart failure,CHF)大鼠心功能及心肌组织代谢物的影响,比较两穴作用机制的差异.方法 在40只无特定病原体(specific pathogen free,SPF)级Sprague-Dawley(SD)大鼠中随机抽取12只作为空白组,将剩余大鼠以腹腔注射盐酸多柔比星法制备CHF大鼠模型.在空白组和造模大鼠中随机抽取 4 只进行模型验证.将造模成功的大鼠随机分为模型组、内关组和膻中组,每组 8 只.内关组和膻中组分别予以电针干预 14 d.观察 4 组大鼠一般行为学、干预结束后超声心动图检测指标、心肌组织病理形态学的变化,用液相色谱-质谱联用(liquid chromatograph-mass spectrometer,LC-MS)检测 4 组大鼠心肌代谢物,用 MetaboAnalyst 6.0 进行代谢组学分析.结果 与空白组比较,模型组大鼠射血分数(ejection fraction,EF)和左室内径缩短率(fractional shortening,FS)下降(P<0.05),光镜下心肌组织明显损伤.与模型组比较,内关组和膻中组 EF和FS 均升高(P<0.05),心肌组织病理损伤减轻.代谢组学结果显示,与空白组比较,模型组大鼠心肌组织中代谢物尿嘧啶、鸟苷、黄嘌呤含量上升(P<0.05),棕榈酰肉碱、腺苷二磷酸葡萄糖、腺苷酸含量下降(P<0.05);与模型组比较,内关组中缬氨酸、谷氨酸、脯氨酸、γ-氨基丁酸、组氨酸、酪氨酸等 16种代谢物含量上升(P<0.05),膻中组中尿苷、谷胱甘肽、6-磷酸果糖、肌酐含量下降(P<0.05);与膻中组比较,内关组腺苷二磷酸核糖酸、牛磺酸、缬氨酸、L-色氨酸、L-脯氨酸、尿苷、谷氨酸、γ-氨基丁酸等含量上升(P<0.05),腺苷、腺嘌呤、苯丙氨酸等含量下降(P<0.05).内关组主要调节氨基酸代谢通路;膻中组主要涉及糖代谢通路,其中嘧啶代谢通路与模型组异常代谢通路相同.结论 电针"内关"和"膻中"均可改善 CHF 大鼠的心功能,减轻心肌损伤,其机制可能是通过改善心肌氨基酸、葡萄糖等相关通路调节心肌能量代谢,"内关"广泛调节作用能够整体调节改善 CHF 病理状态,而"膻中"特异性调节心肌损伤相关代谢物通路,如嘧啶代谢.
Objective To observe the effects of electroacupuncture at"Neiguan"(PC6)and"Danzhong"(CV17)on cardiac function and myocardial tissue metabolites in rats with chronic heart failure(CHF)and investigate the differences in the mechanism of action between the two acupoints.Method Twelve out of 40 specific pathogen-free(SPF)Sprague-Dawley(SD)rats were randomly selected as a control group,and the remaining rats were administered Doxorubicin hydrochloride via intraperitoneal injection to establish CHF models.The control group and the CHF-induced group each randomly contributed 4 rats for model verification.The successfully modeled rats were randomized into a model group,a Neiguan group,and a Danzhong group,with 8 rats in each group.The Neiguan group and Danzhong group received a 14-day electroacupuncture intervention,respectively.General behavioral changes,post-intervention echocardiographic parameters,and myocardial histopathological morphology were observed in the four groups;myocardial metabolites were detected using liquid chromatography-mass spectrometry(LC-MS),and metabolomic analysis was conducted using MetaboAnalyst 6.0.Result Compared with the control group,the ejection fraction(EF)and the left ventricular fractional shortening(FS)were reduced in the model group(P<0.05),along with significantly damaged myocardial tissues under the light microscope.Compared with the model group,the EF and FS were increased in the Neiguan and Danzhong groups(P<0.05),and histopathological damage to myocardial tissues was alleviated.Metabolomics results revealed that,compared with the control group,the model group exhibited increased levels of metabolites uracil,guanosine,and xanthine in the myocardial tissues(P<0.05)and decreased levels of palmitoylcarnitine,adenosine diphosphate glucose,and adenylate(P<0.05).Compared with the model group,the Neiguan group showed elevated contents of 16 metabolites in the myocardial tissues,including valine,glutamate,proline,γ-aminobutyric acid,histidine,and tyrosine(P<0.05),and the Danzhong group demonstrated decreased contents of uridine,glutathione,fructose-6-phosphate,and creatinine(P<0.05).Compared with the Danzhong group,the Neiguan group exhibited increased contents of adenosine diphosphoribose,taurine,valine,L-tryptophan,L-proline,uridine,glutamate,and γ-aminobutyric acid(P<0.05),alongside decreased levels of substances like adenosine,adenine,and phenylalanine(P<0.05).The Neiguan group primarily regulated amino acid metabolic pathways,whereas the Danzhong group mainly involved glucose metabolic pathways,among which the pyrimidine metabolic pathway was an aberrant metabolic pathway observed in the model group.Conclusion Electroacupuncture at"Neiguan"and"Danzhong"can both improve cardiac function and reduce myocardial damage in CHF rats,possibly by regulating myocardial energy metabolism through amino acid and glucose metabolic pathways.The broad regulatory effect of"Neiguan"enables holistic modulation and improvement of the pathological state in CHF,while"Danzhong"specifically targets and regulates metabolic pathways associated with myocardial injury,such as the pyrimidine metabolism.
