Experimental observation on the protective effect of aqueous decoction of Ludiju against acute lung injury in septic rats

目的:观察卤地菊水煎液对脓毒症大鼠急性肺损伤(ALI)的保护作用并探讨其作用机制.方法:将70只雄性SD大鼠按随机数字表分为6组:假手术组、模型组、阳性药组、卤地菊低、中、高剂量组.假手术组和模型组大鼠每日给予蒸馏水10 ml/kg灌胃,给药组分别给予地塞米松(0.05 mg/kg)、卤地菊水煎液(3.15 g/kg,6.30 g/kg,12.0 g/kg)灌胃,1次/d,连续4 d.末次给药后3 h采用盲肠结扎穿刺术(CLP)制备脓毒症ALI模型.术后18 h观察大鼠的死亡率,存活大鼠计算肺湿干重比值(WW/DW),观察肺组织病理形态,检测血清肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-18(IL-18)含量与超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活力,取右肺观察病理形态及检测NLRP3、TXNIP、Caspase-1蛋白表达.结果:与模型组比较,给药各组大鼠死亡率下降,卤地菊高剂量组死亡率最低(33%);给药各组大鼠肺WW/DW 比值均下降,差异均具有统计学意义(P<0.01);给药各组大鼠肺组织中性粒细胞浸润、肺泡壁增厚及实变、肺间质水肿、出血情况减轻.与模型组比较,卤地菊低、中、高剂量组血清TNF-α、IL-1β、IL-18含量下降,阳性药组TNF-α、IL-18含量下降,卤地菊低、高剂量组SOD活力上升,卤地菊中、高剂量组GSH-Px活力上升,阳性药组SOD活力下降、GSH-Px活力上升,阳性药组、卤地菊低、中、高剂量组NLRP3、TXNIP、Caspase-1蛋白表达下降,差异均具有统计学意义(P<0.05,P<0.01).结论:卤地菊水煎液对脓毒症大鼠ALI具有保护作用,其机制可能与抑制炎症反应、增强抗氧化能力、调控TXNIP/NLRP3通路相关蛋白有关.

Objective:To observe the protective effect of the aqueous decoction of Ludiju against acute lung injury(ALI)in septic rats and explore its mechanism of action.Methods:Seventy male Sprague-Dawley rats were randomly di-vided into 6 groups using a random number table:sham operation group,model group,positive drug group,and Ludiju low-dose,medium-dose,and high-dose groups.Rats in the sham operation and model groups were given distilled water(10 ml/kg)by gavage daily.Rats in the administration groups were given dexamethasone(0.05 mg/kg)or Ludiju aqueous decoction(3.15 g/kg,6.30 g/kg,12.0 g/kg)by gavage,once daily for 4 consecutive days.Three hours after the last admin-istration,the sepsis ALI model was established using cecal ligation and puncture(CLP).The mortality rate of the rats was observed 18 hours after surgery.The lung wet/dry weight ratio(WW/DW)of the surviving rats was calculated to observe the pathological morphology of lung tissue.The serum levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),in-terleukin-18(IL-18)and the activity of superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)were detected.The right lung was collected for pathological observation and to detect the protein expression of NLRP3,TXNIP and Caspase-1.Results:Compared with the model group,the mortality rate decreased in all administration groups,with the Ludiju high-dose group having the lowest mortality rate(33%).The lung WW/DW ratio decreased in all administration groups,and the differences were statistically significant(P<0.01).Neutrophil infiltration,alveolar wall thickening and solidation,pulmo-nary interstitial edema and hemorrhage were alleviated in the treated groups..Compared with the model group,serum levels of TNF-α,IL-1βand IL-18 decreased in the Ludiju low-,medium-,and high-dose groups;TNF-α and IL-18 levels de-creased in the positive drug group;SOD activity increased in the Ludiju low-and high-dose groups;GSH-Px activity in-creased in the Ludiju medium-and high-dose groups;SOD activity decreased and GSH-Px activity increased in the posi-tive drug group;and the protein expression of NLRP3,TXNIP and Caspase-1 decreased in the positive drug group and all Ludiju dose groups.The differences compared with the model group were statistically significant(P<0.05,P<0.01).Conclusion:The aqueous decoction of Ludiju has a protective effect on rats with ALI induced by sepsis.Its mechanism may be related to inhibiting the inflammatory response,enhancing antioxidant capacity,and regulating proteins associated with the TXNIP/NLRP3 pathway.
