目的 观察自噬相关基因13(ATG13)蛋白在胃腺癌组织中的表达变化,分析其对患者预后的影响.方法 选择99份胃腺癌原发灶活检标本及71份癌旁组织标本,采用免疫组化法检测ATG13蛋白,比较不同临床病理特征患者胃腺癌ATG13蛋白表达情况,采用Kaplan-Meier曲线比较不同ATG13蛋白表达胃腺癌患者总生存期(OS),采用Cox比例风险回归模型分析胃腺癌患者预后不良的危险因素.结果 与癌旁组织比较,胃腺癌组织ATG13蛋白高表达率高(P<0.05).与ATG13蛋白低表达者比较,ATG13蛋白高表达者胃腺癌肿瘤最大径大(P<0.05),年龄、性别、肿瘤浸润深度、淋巴结转移、远处转移、临床分期及病理分级等指标差异均无统计学意义(P均>0.05).Kap-lan-Meier 生存分析结果提示,与ATG13蛋白低表达者比较,ATG13蛋白高表达胃腺癌患者OS短(P<0.05).单因素Cox回归结果显示,临床分期、淋巴结转移、远处转移及ATG13蛋白表达均与胃腺癌预后不良有关(P均<0.05);多因素Cox回归结果显示,远处转移及ATG13高表达是胃腺癌患者预后不良的危险因素(P均<0.05).结论 胃腺癌组织中ATG13蛋白高表达.ATG13蛋白高表达与胃腺癌患者的预后不良密切相关.

Objective To investigate the expression changes of autophagy-related gene 13(ATG13)protein in gastric adenocarcinoma tissues and to analyze its effect on prognosis of patients.Methods Immunohistochemistry(IHC)was performed to assess ATG13 protein expression in 99 cases of primary gastric adenocarcinoma biopsy tissues and in 71 cases of adjacent tissues.ATG13 protein expression was compared among patients with different clinicopathological characteristics.Kaplan-Meier survival analysis was used to compare overall survival(OS)between patients with high and low ATG13 ex-pression.In addition,a Cox proportional hazards regression model was applied to identify risk factors for poor prognosis in patients with gastric adenocarcinoma.Results The high-expression rate of ATG13 protein was significantly higher in the gastric adenocarcinoma tissues than in the adjacent tissues(P<0.05).Compared with patients with low ATG13 expression,those with high ATG13 expression had a larger maximum tumor diameter(P<0.05).No significant differences were found between the two groups in age,sex,depth of tumor invasion,lymph node metastasis,distant metastasis,clinical stage,or pathological grade(all P>0.05).Kaplan-Meier survival analysis indicated that OS was shorter in patients with high ATG13 expression than in those with low ATG13 expression(P<0.05).Univariate Cox regression analysis showed that clinical stage,lymph node metastasis,distant metastasis,and ATG13 expression were associated with poor prognosis(all P<0.05).Multivariate Cox regression analysis further demonstrated that distant metastasis and high ATG13 expression were in-dependent risk factors for poor prognosis in patients with gastric adenocarcinoma(all P<0.05).Conclusion ATG13 pro-tein is highly expressed in gastric adenocarcinoma tissues,and high ATG13 expression is closely associated with poor prog-nosis in patients with gastric adenocarcinoma.
