高浓度Zebularine诱发小偃麦染色体变异研究OA
Chromosomal Variations in Trititrigia Induced by High Concentration of Zebularine
化学诱变是诱导植物性状变异的重要途径之一,当前最常用的化学诱变剂是甲基磺酸乙酯(EMS),但利用胞嘧啶类似物泽布拉尼(Zebularine)诱导麦类作物选育优异种质资源的研究鲜有报道.本研究以高浓度Zebularine为诱变剂,对3个小偃麦(XY78829、XY7430和XY96253)的萌动种子进行诱导处理,结果表明,与阴性对照(ddH2O)相比,Zebularine处理的小偃麦根尖有丝分裂中期细胞中有丰富的染色体结构变异,主要为染色体易位(429个,占比59.0%)、基因组缺失或断片(298个,占比41.0%).其中基因组间的易位频率(380个,占比52.3%)明显高于基因组内易位(49个,占比6.7%),且基因组间易位发生在A、B基因组之间的最多(111个,15.3%).该结论可为进一步转移和利用偃麦草属有益基因提供理论依据.
Chemical mutagenesis is one of the important approaches to induce plant trait variation.Cur-rently,the most commonly used chemical mutagen is ethyl methyl sulfonate(EMS).However,there are few reports on the use of cytosine analog Zebularine for induction and selection of superior germplasm resources in cereal crops.In this study,high concentration of Zebularine was used as the mutagen to treat germinated seeds of three Trititrigia lines(XY78829,XY7430,and XY96253).The results showed that abundant chromosomal structural variations were observed in the metaphase cells of Trititrigia root tips compared with the negative control(ddH2O),mainly including chromosomal translocations(429,59.0%)and deletions or fragments(298,41.0%).The frequency of intergenomic translocations(380,52.3%)was significantly higher than that of intragenomic translocations(49,6.7%).Within intergenomic translocations,the most translocations oc-curred between homeologous group A and B(111,15.3%).These conclusions could provide valuable infor-mation for further transfer and utilization of beneficial genes from Thinopyrum.
