石笔木不同部位提取物体外降血糖功效的比较OA
Comparison of hypoglycemic effect of extracts from different parts of Tutcheria austrosinica in vitro
目的 探讨石笔木不同部位提取物对α-葡萄糖苷酶的抑制作用.方法 采用回流提取法分别制得石笔木花、花蕊、叶、果皮、种子的乙醇提取物和水提取物,以4-硝基苯基-D-吡喃葡萄糖苷(PNPG)为底物,体外测定10种提取物对α-葡萄糖苷酶活性的抑制作用,通过Lineweaver-Burk作图法确定抑制类型.结果 石笔木花、花蕊、果皮和种子提取物具有一定的α-葡萄糖苷酶抑制活性,其中花蕊醇提物的抑制活性相对较高,其半数抑制浓度(IC50)为1.30 μg·mL-1.抑制动力学研究结果表明,石笔木花、花蕊、果皮和种子提取物对α-葡萄糖苷酶的抑制类型均为混合型抑制.结论 石笔木花、花蕊、果皮和种子提取物具有较好的α-葡萄糖苷酶抑制活性.
Objective To explore the α-glucosidase inhibitory activities of the extracts of different parts of Tutcheria austrosinica,the ethanol extracts and water extracts of the flowers,stamens,leaves,pericarps and seeds of T.austrosinica were prepared by refluxing extraction.Methods The α-glucosidase inhibitory activities of 10 kinds of extracts were screened with 4-nitrophenyl-D-glucopyranoside(PNPG)as substrate,the kinetics of active extracts was studied by enzyme-catalyzed reaction,and the Lineweaver-Burk curve was drawn to deduce their inhibitory types.Results The extracts of flowers,stamens,pericarps and seeds showed potent α-glucosidase inhibitory activities,and the inhibitory activity of the extract from the stamens was relatively high,with an IC50 value of 1.30 μg·mL-1.Inhibition dynamics analysis showed that the extracts of flowers,stamens,pericarps and seeds were all close to mixed modes of inhibition against α-glucosidase.Conclusion The extracts of flowers,stamens,pericarps and seeds of T.austrosinica exhibited promising α-glucosidase inhibitory activities.
