牛蒡根总多酚提取工艺以及含量测定OA
Study on optimization of extraction technology and accurate content determination of total polyphenols from Arctium lappa L.root
目的 优化牛蒡根总多酚的提取工艺,建立纯化富集方法并测定其特征指标成分含量,为后续研究提供基础.方法 以总多酚含量为评价指标,通过单因素试验筛选乙醇体积分数、料液比、提取温度的最佳参数;经石油醚脱脂、正丁醇萃取及AB-8型大孔吸附树脂富集纯化提取物;采用福林-酚法测定总多酚含量,高效液相色谱(HPLC)法定量7种指标性多酚成分.结果 最佳提取工艺为70%乙醇、料液比1∶15(g∶mL)、60℃提取,粗提物总多酚含量2.71%;经纯化富集后,总多酚含量提升至59.84%.HPLC测定显示,新绿原酸、绿原酸等7种指标成分总占比达提取物的28.05%.结论 本研究确定的提取与富集工艺高效可行,明确了提取物的主要成分含量,为牛蒡根多酚的开发利用提供了技术支撑.
Objective To optimize the extraction process of total polyphenols from Arctium lappa L.roots,establish a purification and enrichment method,and determine the content of its characteristic marker components,so as to provide a basis for subsequent research.Methods With total polyphenol content(TPC)as the evaluation index,single-factor experiments were conducted to screen the optimal parameters of ethanol concentration,solid-to-liquid ratio,and extraction temperature.The crude extract was purified and enriched by sequential petroleum ether defatting,n-butanol extraction,and AB-8 type macroporous adsorption resin.Folin-Ciocalteu method was used to determine the TPC,and high-performance liquid chromatography(HPLC)was applied for the quantitative analysis of 7 marker phenolic acid components.Results The optimal extraction conditions were 70%ethanol,a solid-to-liquid ratio of 1∶15(g∶mL),and extraction at 60 ℃,under which the TPC of the crude extract reached 2.71%.After purification and enrichment,the TPC was significantly increased to 59.84%.HPLC analysis showed that the total content of 7 marker components(including neochlorogenic acid and chlorogenic acid)accounted for 28.05%of the purified extract.Conclusion The established extraction and enrichment processes are efficient and feasible,and the main component contents of the extract are clarified,which provides technical support for the development and utilization of Arctium lappa L.root polyphenols.
