Study on evaluating overall quality consistency of red yeast rice using Bi-value and Tri-spectrum method

目的 建立红曲质量标准数字化控制方法,形成红曲质量标准的数字化控制新判据标准.方法 用二值三谱法及估纯二值法综合评价鉴定红曲整体质量均一性和批次质量的一致性.采用C18色谱柱(4.6 mm×250 mm,5 μm),以乙腈-0.05%磷酸梯度洗脱,检测波长237、256 nm,流速1.0 mL·min-1,进样量20 μL,柱温30 ℃.结果 以染料木素(genistein,GEN)为参照物峰,确认10个共有指纹峰,方法学验证显示精密度、重复性和12 h内供试液稳定性的相对标准偏差(RSD)均小于3.0%,符合中药指纹图谱技术要求.建立256 nm-高效液相色谱(HPLC)红曲指纹图谱,以二值三谱法鉴别31批红曲宏定性相似度Sm均>0.97,而宏定量相似度Pm波动范围为76.7%~124.7%,将其分为5级,除5级样品(S2、S6、S8、S9、S16、S30)外均合格.对标准曲线法和三谱定量法(即比率指纹定量,R-SFP法)的三指标含量结果进行对比,结果显示二者具有高度一致性,二值三谱法可作为复杂中药整体质量控制和评价的简捷方法.建立237、256 nm双波长吸光系数比谱,通过估纯二值法对红曲10个指纹峰进行整体鉴定分析,全部指纹峰纯度合格.结论 二值三谱法能有效评价和测定红曲的10个特征指纹物质总含量及分含量,估纯二值法可一次性整体鉴别全部指纹峰纯度,用数字化质量标准控制中药质量简便可行.

Objective To establish a digitized control method for the quality standard of red yeast rice,for which serves as a novel evaluating criteria for its quality standard.Methods The overall quality uniformity of red yeast rice and batch-to-batch consistency were comprehensively evaluated using Bi-value and Tri-spectrum method(BVTSM)and estimating purity Bi-value method(EP-BVM).Separation was performed on a C18 column(4.6 mm×250 mum,5 μm)using a gradient elution of acetonitrile and 0.05%phosphoric acid.Detection wavelengths were set at 237 nm and 256 nm,with a flow rate of 1.0 mL·min-1,injection volume of 20 μL,and column temperature of 30 ℃.Results Using genistein as the reference peak,the 10 common fingerprint peaks were analyzed.Methodology validation showed that the Relative standard deviation(RSD)values for precision,repeatability,and sample solution stability tests within 12 hours were all less than 3.0%,for which meet the technical requirements for traditional Chinese medicine(TCM)fingerprint.The 256 nm-HPLC fingerprint of red yeast rice was established.Using BVTSM,the macro-qualitative similarity Sm of 31 batches of red yeast rice samples were all>0.97,while the macro-quantitative similarity Pm fluctuated from 76.7%to 124.7%.Samples were classified into five grades,with all batches qualifying except those in grade 5(S2,S6,S8,S9,S16,S30).The contents from the standard curve method and the ratio fingerprint quantitation(R-SFP)Method for three indicators were compared,showing high consistency.BVTSM can serve as a concise approach for the overall quality control and evaluation of complex TCMs.The dual-wavelength absorbance ratio spectrum was established,and the overall purity of all 10 fingerprint peaks of red yeast rice was identified and analyzed using the EP-BVM,confirming that all the fingerprint peaks met purity standards.Conclusion BVTSM can effectively evaluates and determines the total pharmacologically active substance content in red yeast rice,while the EP-BVM can comprehensively identify the purity of all fingerprint peaks at one time.Digitized HPLC fingerprint is an accurate and feasible approach for controlling TCM quality standard.
