趾腿病鸵鸟主要器官的组织病理学及骨免疫相关基因在骨-免疫器官中表达研究OA
Histopathology Study of Main Organs and Expression of Bone Immune-Related Genes in Ostriches with Toe and Leg Disease
[目的]研究趾腿病鸵鸟主要器官的组织病理学及骨免疫相关基因在跗关节与免疫器官中的表达变化,为探明趾腿病鸵鸟的发病机制及防治提供理论依据.[方法]以健康、趾腿病雄性雏鸵鸟为研究对象,通过大体解剖、病理组织学研究鸵鸟的临床特征部位(跗关节、肌肉)、免疫器官(脾脏、胸腺、法氏囊、哈德氏腺)、内脏(心脏、肝脏、肺脏、肾脏)、肠(十二指肠、空肠、回肠、盲肠、结直肠)的组织病理学变化;对临床特征部位跗关节软骨进行转录组分析,筛选出IL-1R2、MMP7、MHC Ⅱ、ATF3、CSF3R、LAPTM4B、LTA4H、MLKL骨免疫相关的差异表达基因,并进行qPCR验证;研究以上8个骨免疫相关基因在跗关节与免疫器官中的表达规律.[结果]①临床及大体解剖观察:趾腿病鸵鸟跗关节的关节软骨结构模糊,关节切面有黄色液体渗出.②病理组织学观察:跗关节软骨细胞减少、坏死,并伴较多空软骨陷窝,肌纤维萎缩;脾脏、胸腺可见较多坏死细胞,法氏囊淋巴细胞减少;心脏心肌细胞脂肪变性;肝脏中央静脉淤血;肺间质组织增多、肺泡壁增厚,肺泡间质可见炎性细胞浸润;肾脏未见明显病理变化;除空肠中杯状细胞减少外,其他肠道未见明显变化.③转录组分析:与鸵鸟参考基因组比对,共有29个DEGs;GO分析上述DEGs,其集中在初级代谢过程、免疫系统过程等生物学过程;KEGG pathway分析显示多条通路均与免疫相关,尤其与T细胞增殖活化相关.④ qPCR结果显示:以上8个骨免疫相关基因在健康鸵鸟的跗关节、免疫器官中均有表达;与健康鸵鸟相比,其在趾腿病鸵鸟跗关节软骨中均升高;IL-1R2、MHC Ⅱ、ATF3、CSF3R、LAPTM4B在胸腺与法氏囊均显著(P<0.05)升高,MMP7在脾脏中极显著(P<0.01)下降、在法氏囊中显著(P<0.05)升高.[结论]趾腿病鸵鸟除关节病变外还伴有多个器官病变,其中胸腺、脾脏、法氏囊病变最为严重;GO与KEGG结果显示免疫系统及与免疫相关通路参与其中;骨免疫相关基因在趾腿病鸵鸟跗关节与免疫器官中显著变化.以上提示8个骨免疫相关基因参与了趾腿病鸵鸟跗关节与免疫器官的病理损伤.
[Objective]This study aimed to investigate the histopathological changes in major organs and the expression of bone immune-related genes in the tarsal joint and immune organs of ostrich with toe and leg disease,thereby providing a theoretical basis for elucidating the pathogenesis and prevention of os-trich with toe and leg disease.[Method]Healthy and diseased male ostrich chicks were used as re-search subjects.The histopathological changes of clinical characteristic parts(tarsal joints,muscles),immune organs(spleen,thymus,bursa of Fabricius,Hadrian's gland),visceral organs(heart,liver,lungs,kidneys),and intestines(duodenum,jejunum,ileum,cecum,colon and rectum)of ostriches were studied by gross dissection and histopathology;transcriptome analysis was conducted on tarsal joint cartilage to screen for differentially expressed bone immunity-related genes,such as IL-1R2,MMP7,MHC Ⅱ,ATF3,CSF3R,LAPTM4B,LTA4H,and MLKL,followed by qPCR validation;the expression patterns of these eight bone immune-related genes in the tarsal joint and immune organs were studied.[Result]① Clinical and gross anatomical observations showed blurred cartilage structure in the tarsal joint of affected ostriches,with yellow fluid seeps out from the joint section.② Histopathological exami-nation revealed decreased and necrosis chondrocytes in the tarsal joint,accompanied by numerous empty cartilage pits and muscle fiber atrophy.Increased necrotic cells were observed in the spleen and thymus,while lymphocyte numbers were reduced in the bursa of Fabricius.The heart showed fatty degeneration of cardiomyocytes.The liver showed central venous congestion.The lungs presented increased interstitial tis-sue,thickened alveolar walls,and inflammatory cell infiltration within the alveolar interstitium.No sig-nificant pathological changes were observed in the kidneys.Apart from a decrease in goblet cells in the je-junum,other intestinal segments showed no notable alterations.③ Transcriptome analysis identified 29 differentially expressed genes(DEGs)compared to the ostrich reference genome.GO analysis revealed that these DEGs were enriched in biological processes such as primary metabolism and immune system processes.KEGG pathway analysis revealed that several pathways were associated with immunity,par-ticularly those related to T-cell proliferation and activation.④ The qPCR results indicated that the eight bone immune-related genes were expressed in the tarsal joints and immune organs of healthy ostriches,and were upregulated in the tarsal cartilage of diseased ostriches.IL-1R2,MHC Ⅱ,ATF3,CSF3R,and LAPTM4B were significantly upregulated(P<0.05)in both the thymus and bursa of Fabri-cius.MMP7 expression was significantly decreased(P<0.01)in the spleen and significantly increased(P<0.05)in the bursa of Fabricius.[Conclusion]In addition to joint lesions,ostriches with toe and leg disease exhibited multiple organ lesions,with the thymus,spleen,and bursa of Fabricius being most severely affected.GO and KEGG results indicated the involvement of immune system processes and related pathways.Bone immunity-related genes showed significant expression changes in the tarsal joint and immune organs of affected ostriches.These findings suggest that the eight bone immunity-related genes are involved in the pathogenesis of tarsal joint and immune organ damage in ostriches with toe and leg disease.
