有色玉米籽粒花青素成分解析及分类方法建立OA

[目的]旨在构建一套有色玉米籽粒分类方法,构建定量化分类体系以提升花青素检测效率与品种筛选精度.[方法]研究以212份富含花青素的纯合玉米自交系为材料,通过目测法与比色卡进行籽粒颜色表型观察,并利用高效液相色谱技术对花青素成分进行定性与定量分析,以精确探究颜色特征与花青素组成之间的相关性.[结果]有色甜、糯玉米自交系籽粒花青素核心组分均为矢车菊色素和天竺葵色素,甜玉米总花青素含量718.38 mg/kg,较糯玉米高1.07倍,二者在芍药素检出率、矢车菊色素贡献率上差异显著;云南种植点花青素含量较四川提升8.31%,筛选出25份高花青素种质材料,其中糯玉米24CH92含量达2 884.09 mg/kg;将212份材料分为48类,PANTONE 229 U类群花青素含量最高723.43 mg/kg,且RGB值与花青素呈极显著负相关.[结论]建立了基于籽粒颜色RGB模型的高效、低成本花青素含量预测体系,明确了籽粒颜色与花青素组成的定量关系,为有色鲜食玉米的精准育种和种质创新提供了实用化技术路径.

[Objective]This study aims to establish a classification system for pigmented maize kernels and develop a quantitative classification framework to improve the efficiency of anthocyanin detection and the accuracy of variety screening.[Method]A total of 212 anthocyanin-rich homozygous maize inbred lines were used as materials.Kernel color phenotypes were observed via visual inspection and color cards,while qualitative and quantitative analyses of anthocyanin components were performed using high-performance liquid chromatography(HPLC)to precisely investigate the correlation between color charac-teristics and anthocyanin composition.[Result]The core anthocyanin components in the kernels of pig-mented sweet and waxy maize inbred lines were cyanidin and pelargonidin.The total anthocyanin content in sweet maize was 718.38 mg/kg,which was 1.07 times higher than that in waxy maize.Significant differ-ences were observed between the two types in the detection rate of peonidin and the contribution rate of cyanidin.The anthocyanin content in kernels grown in Yunnan was 8.31%higher than that in Sichuan.A total of 25 germplasm materials with high anthocyanin content were selected,among which the waxy maize line 24CH92 exhibited the highest content at 2,884.09 mg/kg.The 212 inbred lines were classified into 48 groups.The PANTONE 229 U group showed the highest anthocyanin content at 723.43 mg/kg,and RGB values were significantly negatively correlated with anthocyanin content.[Conclusion]An effi-cient and low-cost prediction system for anthocyanin content was established based on the RGB model of kernel color.The quantitative relationship between kernel color and anthocyanin composition was clari-fied,providing a practical technical pathway for the precision breeding and germplasm innovation of pig-mented fresh-eating maize.
