[目的]旨在研究提升羊边界病毒(border disease virus,BDV)的快速检测方法.[方法]基于BDV保守区域5'UTR序列设计特异性引物,通过优化反应体系参数(包括退火温度、引物浓度及纳米颗粒添加比例),成功构建BDV纳米PCR(Nano-PCR)检测方法.对比分析了其灵敏度、特异性及临床适用性,并与普通PCR方法进行平行验证.[结果]优化后的Nano-PCR可稳定扩增出大小为210 bp的BDV特异性靶标,其最低检测限为1.1×10 copies/μL,灵敏度较普通PCR(1.1×103 copies/μL)提升100倍.在特异性检测中,BDV纳米PCR检测方法对牛病毒性腹泻病毒、小反刍兽疫病毒及羊肠道病毒均无交叉反应.对50份临床样品的检测显示,Nano-PCR阳性检出率为6%(3/50),与普通PCR结果完全一致,但灵敏度显著高于普通PCR.[结论]建立的BDV纳米PCR检测方法较普通PCR具有更高灵敏度和特异性,可为羊群边界病毒感染的早期诊断及流行病学监控提供技术支持.

[Objective]The aim is to develop a rapid detection method for border disease virus(BDV).[Method]Specific primers were designed based on the conserved 5'UTR sequence of BDV.By optimizing the reaction system parameters(including annealing temperature,primer concentration,and nanoparticle addition ratio),a BDV Nano-PCR detection method was successfully constructed.The sensitivity,specificity,and clinical applicability of this method were compared and analyzed and a parallel verification was conducted with those of the conventional PCR method.[Result]The optimized Nano-PCR could stably amplify a BDV-specific target of 210 bp,with a minimum detection limit of 1.1×10 copies/μL,and its sensitivity was 100 times higher than that of the conventional PCR(1.1×103 copies/μL).In the specific detection,this method showed no cross-reaction with bovine viral diarrhea virus,peste des petits ruminants virus,and Caprine enterovirus.The detection of 50 clinical samples showed that the positive detection rate of Nano-PCR was 6%(3/50),which was completely consistent with the results of the conventional PCR,but the sensitivity was significantly higher than that of the conventional PCR.[Conclusion]the established BDV Nano-PCR detection method has higher sensitivity and specificity than the conventional PCR,and can provide technical support for the early diagnosis and epidemiological monitoring of border disease virus infections.
