目的 探讨肺不藏魄不寐大鼠相关脏器中Bcl-2、Bax、Caspase-3表达水平,为中医不寐五神分型提供基础实验依据.方法 按照随机数字表法将16只SPF级雄性SD大鼠分为大平台对照组及肺不藏魄不寐组,每组8只.大平台对照组采用水环境大平台法,肺不藏魄不寐组采用水环境小平台法建立模型.监测两组大鼠一般情况并进行水迷宫实验.采用HE观察两组大鼠肺组织病理变化,采用IHC及WB检测两组大鼠脑组织及肺组织中Caspase-3、Bcl-2、Bax表达水平.结果 与大平台对照组相比,一般情况显示:肺不藏魄不寐组大鼠造模后精神逐渐萎靡,毛发暗淡、时有脱毛,饮食逐渐变差;水迷宫结果示:肺不藏魄不寐组第二目标象限滞留时间、进入次数、目标象限运动距离及找到目标靶平台运动轨迹增加(P<0.05);HE染色:肺不藏魄不寐组肺组织显示肺泡结构排列紊乱,肺泡上皮细胞水肿,肺泡间隔增宽,见较多淋巴细胞和浆细胞浸润;IHC显示:肺不藏魄不寐组脑组织及肺组织Bcl-2表达减少(P<0.05),Bax和Caspase-3表达增加(P<0.05);WB结果示:肺不藏魄不寐组脑组织及肺组织Bcl-2表达显著减少(P<0.01),Bax及Caspase-3表达增加(P<0.05).结论 脑组织及肺组织凋亡水平的改变与肺不藏魄不寐的发生有关.

Objective To investigate the expression levels of Bcl-2,Bax and Caspase-3 in the related organs of insomnia rats with lung not storing corporeal soul,so as to provide a basic experimental basis for the traditional Chinese medicine(TCM)classification of the five spirits of insomnia.Methods According to the random number table method,16 SPF male SD rats were di-vided into large platform control group and lung not storing corporeal soul group,with 8 rats in each group.The large platform con-trol group was used to establish the water environment large platform method,and the lung not storing corporeal soul group was used to establish the model by the water environment small platform method.The general condition of the rats in the two groups was monitored and the water maze experiment was performed.HE was used to observe the pathological changes of lung tissues of the two groups,and immunohistochemistry(IHC)method and Western Blot(WB)were used to detect the expression levels of Caspase-3,Bcl-2 and Bax in the brain and lung tissues of the two groups.Results Compared with those in the control group,the general situation showed that the rats in the lung not storing corporeal soul group gradually became depressed,their hair was dull,there was hair loss from time to time,and their diet gradually deteriorated(P<0.05).HE staining showed disordered alveolar structure,alveolar epithelial cell edema,widened alveolar septum and more lymphocyte and plasma cell infiltrates.IHC method showed that the expression of Bel-2 in brain tissue and lung tissue decreased(P<0.05),and the expressions of Bax and Caspase-3 increased(P<0.05).WB results showed that the expression of Bcl-2 in brain tissue and lung tissue in lung tissue was significantly decreased(P<0.01),the expressions of Bax and Caspase-3 increased(P<0.05).Conclusion The changes of apoptosis in brain and lung tissues are related to the occurrence of lung not storing corporeal soul.
