目的:构建一种兼具三维多孔结构、生物活性信号及抗菌功能的负载S-亚硝基-N-乙酰基青霉胺(S-ni-troso-N-acetyl-penicillamine,SNAP)的 3D打印支架,以实现高效骨再生并防控术后感染.方法:采用数字光处理(dig-ital light processing,DLP)技术制备水凝胶支架,并负载一氧化氮(NO)供体SNAP,表征支架形貌及NO释放;大鼠骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)及MC3T3-E1 细胞评价支架生物相容性及体外成骨分化能力,用平板涂布法检测支架抑菌作用.结果:制备了负载SNAP的 3D打印支架,可持续释放NO至少 3 d,相较于对照组,能有效抑制致病菌生长、上调成骨基因表达.结论:负载SNAP的 3D打印支架可持续释放NO,同时实现有效抗菌与促成骨分化,兼具骨修复与感染防护功能,具良好骨修复应用前景.

Objective:To develop a S-nitroso-N-acetyl-penicillamine(SNAP)loaded 3D printed scaffold integrating a three-dimensional porous architecture,bioactive signaling,and antibacterial properties,aiming to achieve efficient bone regen-eration while preventing postoperative infections.Methods:Hydrogel scaffolds were fabricated utilizing digital light processing(DLP)technology and loaded with nitric oxide(NO)donor,SNAP.Scaffold morphology and NO release kinetics were subse-quently characterized.Bone marrow mesenchymal stem cells(BMSCs)and MC3T3-E1 cells were employed to evaluate the biocompatibility and in vitro pro-osteogenic capacity of the scaffolds.Furthermore,the antibacterial efficacy of the scaffolds was assessed using the agar plate spreading method.Results:The successfully fabricated SNAP loaded 3D printed scaffolds exhibited a sustained release of NO for at least 3 days.Compared to the control group,these scaffolds effectively inhibited the proliferation of pathogenic bacteria and significantly upregulated the expression of osteogenic genes.Conclusion:This study demonstrates that the SNAP-loaded 3D printed scaffold provides sustained NO release,facilitating simultaneous antibacterial and pro-osteogenic effects.By integrating bone repair capabilities with infection prophylaxis,this dual-functional scaffold holds promising clinical potential for application in bone regeneration.
