用于口腔微创治疗的可注射抗菌黏附复合水凝胶的制备及研究OA
Fabrication and study of an injectable antibacterial adhesive composite hydrogel for oral minimally invasive therapy
目的:研制一种用于口腔微创治疗的多功能可注射水凝胶,并评估其性能.方法:构建聚多巴胺修饰纤维素纳米晶/多巴胺接枝透明质酸(polydopamine-modified cellulose nanocrystal/dopamine-drafted hyaluronic acid,PCNC/HA-DA)水凝胶.采用流变学测试其剪切稀化与自愈合性能;通过CCK-8与活死染色检测其对人牙周膜细胞的生物相容性;通过平板计数与结晶紫染色评价其对变异链球菌和粪肠球菌的抗菌及抗生物膜能力.结果:该水凝胶具有显著的剪切稀化与快速自愈合能力,可顺利注射.细胞实验显示材料生物相容性良好.其对两种口腔致病菌均有显著抑制作用,并能有效抑制生物膜形成.结论:PCNC/HA-DA水凝胶兼具可注射性、湿态黏附性、抗菌活性及良好细胞亲和性,在口腔微创治疗中具有应用潜力.
Objective:To develop a multifunctional injectable hydrogel for oral minimally invasive therapy and evalu-ate its properties.Methods:A polydopamine-modified cellulose nanocrystal/dopamine-drafted hyaluronic acid(PCNC/HA-DA)hydrogel was constructed.Its shear-thinning and self-healing properties were assessed via rheological tests,biocompati-bility with human periodontal ligament cells was examined using CCK-8 assay and live/dead staining,antibacterial and anti-biofilm activities against Streptococcus mutans and Enterococcus faecalis were evaluated by plate counting and crystal violet staining.Results:The hydrogel exhibited significant shear-thinning and rapid self-healing capabilities,allowing smooth injec-tion.Cell experiments confirmed favorable biocompatibility.It showed significant inhibitory effects against both pathogens and effectively suppressed biofilm formation.Conclusion:The PCNC/HA-DA hydrogel integrates injectability,wet-adhesion,anti-bacterial activity,and favorable cytocompatibility,demonstrating potential for oral minimally invasive applications.
