在合成木质素脱氢聚合物(DHP)的基础上,以含有DHP的环戊酮为油相,去离子水为水相,纤维素纳米晶体(CNC)做固体颗粒稳定剂,制备了水包油(O/W)型Pickering乳液,以Pickering乳液体系为构建基础,开发了一种新型的药物递送系统.利用傅里叶红外光谱(FT-IR)仪、激光光散射凝胶色谱联用(SEC-MALS)仪、马尔文激光粒度仪和紫外分光光度计等设备,对Pickering乳液的粒径、稳定性及流变特性进行了全面表征.研究结果表明:乳液粒径为430 nm,展现出良好的稳定性.Pickering乳液的载药量为24.7%±2.0%,包封率为80.0%±1.8%.体外实验中,含DHP的CNC-Pickering乳液在人工胃液中表现出良好的稳定性,而在人工结肠液中会出现水油分层的破乳现象.采用噻唑蓝(MTT)比色法对破乳后的DHP进行抗肿瘤活性分析,其对结肠癌HCT116细胞有显著的抑制效果,半数抑制质量浓度(IC50)为186.97 mg/L.木质素模型物/CNC的Pickering乳液可以在酶响应的条件下实现靶向释放,以达到治疗结肠癌的效果.

Based on the synthesis of lignin dehydrogenation polymer(DHP),an oil-in-water(O/W)Pickering emulsion was prepared using cyclopentanone containing DHP as the oil phase,deionized water as the aqueous phase,and cellulose nanocrystals(CNC)as solid particle stabilizers.Using the Pickering emulsion,a novel drug delivery system was developed.The particle size,stability,and rheological properties of the Pickering emulsion were comprehensively characterized with Fourier-transform infrared spectroscopy(FT-IR),size-exclusion chromatography combined with multi-angle light scattering(SEC-MALS),Malvern laser particle size analyzer,and ultraviolet-visible spectrophotometry.Results showed that the emulsion had an average particle size of approximately 430 nm and exhibited good stability.The drug loading capacity of the Pickering emulsion was 24.7%±2.0%,with an encapsulation efficiency of 80.0%±1.8%.In vitro experiments demonstrated that the DHP-containing CNC-Pickering emulsion remained stable in simulated lumen juice but underwent phase separation(i.e.,emulsion breakdown)in simulated colonic juice.After breakdown,the released DHP was evaluated for antitumor activity using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)colorimetric assay.The results indicated significant inhibitory effects against human colon cancer HCT116 cells,with a half-maximal inhibitory concentration(IC50)of 186.97 mg/L.Thus,the lignin model compounds/CNC-based Pickering emulsion enables targeted drug release under enzyme-responsive conditions to achieve the therapeutic effect on colon cancer.
