基于UHPLC-Q-Exactive-Orbitrap MS/MS和网络药理学探讨金线莲抗抑郁有效成分及作用机制研究OA
The Effective Ingredients and Mechanism of Anoectochilus Roxburghii in Treating Depression Based on UHPLC-Q-Exactive-Orbitrap MS and Network Pharmacology
目的:探讨金线莲治疗抑郁症的药效物质基础与潜在作用机制.方法:采用超高效液相色谱-四级杆-静电场轨道阱高分辨质谱(UHPLC-Q-Exactive-Orbitrap MS)分析金线莲水提物的化学成分及入血成分,利用 PubChem、Swiss Target Prediction 数据库获取入血成分的作用靶点;GeneCards、Therapeutic Target、OMIM 数据库获得疾病的作用靶点;运用STRING 12.0 平台构建蛋白-蛋白相互作用 PPI 网络;通过 DAVID 数据库对交集靶点进行基因本体 GO 功能及京都基因与基因组百科全书行 KEGG 通路富集分析.结果:从金线莲水提物中共鉴定包括 42 种化合物,经灌胃后大鼠血浆中鉴定出 14种入血成分.疾病与入血成分交集靶点 265 个,其中关键靶点 53 个,主要涉及 IL-6、AKT1、SRC、STAT3、PPARG等靶点;GO 功能富集和 KEGG 信号通路分析显示金线莲可能通过癌症中的通路、炎症介质对 TRP通道的调节、神经活性配体-受体相互作用、5-羟色胺能突触、VEGF 信号通路、HIF-1 信号通路、PI3K-Akt信号通路、FoxO 信号通路等发挥治疗抑郁症的作用.结论:金线莲中的活性成分金线莲苷、芹菜素、5-羟甲基糠醛等,可能是通过调癌症中的通路、炎症介质对 TRP通道的调节、神经活性配体-受体相互作用、5-羟色胺能突触、VEGF 信号通路、HIF-1 信号通路、PI3K-Akt 信号通路、FoxO信号通路等信号通路及IL-6、AKT1、SRC、STAT3等基因发挥治疗抑郁症的作用.
Objective:To explore the pharmacological material basis and potential mechanism of action of Anoectochilus roxburghii in the treatment of depression.Methods:Ultra-high performance liquid chromatography-quadrupole-Orbitrap high-resolution mass spectrometry(UHPLC-Q-Exactive-Orbitrap MS)was used to analyze the chemical components and blood components of Anoectochilus roxburghii water extract.The target sites of the blood components were obtained from the PubChem and Swiss Target Prediction databases.The target sites of the disease were obtained from the GeneCards,Therapeutic Target,and OMIM databases.The protein-protein interaction(PPI)network was constructed using the STRING platform.The intersection targets were analyzed for gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment using the DAVID database.Results:A total of 42 compounds were identified in the water extract of Anoectochilus roxburghii,and 14 blood components were identified in the plasma of rats after intragastric administration.There were 265 intersection targets between the disease and the blood components,among which 53 were key targets,mainly involving IL-6,AKT1,SRC,STAT3,PPARG,etc.GO functional enrichment and KEGG pathway analysis showed that Anoectochilus roxburghii may exert its therapeutic effect on depression through pathways such as cancer,regulation of TRP channels by inflammatory mediators,neuroactive ligand-receptor interaction,5-hydroxytryptamine(serotonin)synaptic signaling pathway,VEGF signaling pathway,HIF-1 signaling pathway,PI3K-Akt signaling pathway,and FoxO signaling pathway.Conclusion:The active components such as anoectochiluside,apigenin,and 5-hydroxymethylfurfural in Anoectochilus roxburghii may exert its therapeutic effect on depression through pathways such as cancer,regulation of TRP channels by inflammatory mediators,neuroactive ligand-receptor interaction,5-hydroxytryptamine(serotonin)synaptic signaling,VEGF signaling,HIF-1 signaling,PI3K-Akt signaling,and FoxO signaling,as well asgenes such as IL-6,AKT1,SRC,and STAT3.
