目的 观察实炎方对溃疡性结肠炎(UC)小鼠肠道菌群和Th17细胞的影响,初步探索实炎方治疗UC的作用机制.方法 48只小鼠随机分为空白组、模型组、美沙拉秦组和低剂量实炎方组、中剂量实炎方组和高剂量实炎方组,每组8只.模型组给予3%DSS溶液自由饮食造模,低剂量实炎方组、中剂量实炎方组、高剂量实炎方组和美沙拉秦组给予3%DSS溶液自由饮7 d,然后改为无菌水自由饮7 d,且整个过程小鼠予以15、30、60 g/kg实炎方或0.5 g/kg美沙拉秦灌胃.记录小鼠DAI评分,小鼠结肠长度,HE、MASSON染色观察实炎方对UC小鼠肠黏膜炎症影响并进行评分.16S rRNA测序分析实炎方治疗后小鼠肠道菌群的变化,流式细胞术测定小鼠血清中Th17的细胞比例.结果 与模型组相比,实炎方干预后小鼠DAI评分明显降低,结肠长度长于模型组并趋于正常,HE评分、MASSON染色评分也明显低于模型组.实炎方干预后小鼠肠道菌群结构发生明显变化,主要以杆菌纲、乳酸杆菌目、乳酸杆菌属、乳酸杆菌科等为特征性菌群.实炎方干预后小鼠Th17细胞含量也明显低于模型组.结论 实炎方可以通过调节肠道菌群结构,增加乳酸杆菌等有益菌的含量,降低Th17细胞的激化和活性,改善肠道的免疫平衡,缓解肠道炎症.

Objective To observe the effects of Shiyan Decoction on intestinal microbiota and Th17 cells in mice with ulcerative colitis(UC),and to explore its mechanism in the treatment of UC.Methods A total of 48 mice were randomly divided into a blank group,a model group,a mesalazine group,a low-dose Shiyan Decoction group,a medium dose Shiyan Decoction group,and a high-dose Shiyan Decoction group,with 8 mice in each group.The model group was given 3%DSS solution for free diet modeling,while the low-dose Shiyan Decoction group,medium dose Shiyan Decoction group,high-dose Shiyan Decoction group,and mesalazine group were given 3%DSS solution for free drinking for 7 days,and then switched to sterile water for 7 days.During the entire process,the mice were given 15g/kg Shiyan Decoction,30g/kg Shiyan Decoction,60g/kg Shiyan Decoction,or 0.5g/kg mesalazine by gavage.The DAI scores,colon length were recorded,and HE and MASS staining were used to observe the effects of Shiyan Decoction on intestinal mucosal inflammation in UC mice and score them.16S rRNA sequencing was used to analyze the changes in gut microbiota of mice treated with Shiyan Decoction,and flow cytometry was used to determine the proportion of Th17 cells in mouse serum.Results Compared with the model group,after the intervention with Shiyan Decoction,the DAI scores of mice were significantly reduced,and the colon length was longer than that of the model group and tended to be normal,and the HE scores and MASSON staining scores were also significantly lower than those of the model group.After the intervention with Shiyan Decoction,there exhibited significant changes in the gut microbiota structure of mice,mainly characterized by the phylum Bacteroidetes,order Lactobacillales,genus Lactobacillaceae,family Lactobacillaceae,and phylum Firmicutes.After the intervention with Shiyan Decoction,the content of Th17 cells in mice was significantly lower than that in the model group.Conclusion Shiyan Decoction can improve intestinal immune balance and alleviate intestinal inflammation by regulating the structure of intestinal microbiota,increasing the content of beneficial bacteria such as lactobacilli,and reducing the activation and activity of Th17 cells.
