为研究樟子松(Pinus sylvestris var.mongolica)根系分离的暗隔内生真菌(Dark septate endophytes,DSE)对重金属镉(Cd)的耐受性,通过平板与液体发酵培养筛选获得1株耐镉优良菌株A041(Paraphoma chrysanthe-micola),测定不同浓度Cd胁迫下该菌株抗性生理指标,通过扫描电镜观察其菌丝形态变化,并研究其耐(镉)毒性机制.结果表明:在不同浓度Cd胁迫条件下,菌株A041的生长速率、生物量均显著高于其他菌株,该菌株的主要抗性生理指标同对照相比均显著提高,不同处理间差异显著(P<0.05),随着Cd浓度的增加,过氧化氢酶(CAT)、过氧化物酶(POD)和超氧化物歧化酶(SOD)活性分别在Cd浓度为3,5,6 mmol/L时最高,与对照相比分别增加215%,92%,122%,在Cd浓度为7 mmol/L时丙二醛(MDA)和可溶性蛋白含量最高,与对照相比分别增加193%和47%,谷胱甘肽(GSH)含量先增加后降低,在Cd浓度为2 mmol/L时最高,占对照相比增加42%;A041菌丝中黑色素含量随着Cd胁迫的增强呈先增加后减少的趋势,在Cd浓度为2 mmol/L时含量最高,与对照相比增加34%.不同浓度Cd胁迫可改变菌株A041的菌丝形态,Cd浓度增大,菌丝直径减小并且越来越稀疏.综上,菌株A041(P.chrysanthemicola)对镉具有较强的耐受能力.

To explore the tolerance of dark septate endophytes(DSE)in the roots of Pinus sylvestris var.mongolica to heavy metal Cd,we selected a strain with the strongest resistance by plate and liq-uid fermentation culture method,determined its physiological index of resistance under different con-centrations of Cd stress,and observed the morphological changes of mycelia by scanning electron mi-croscopy to further study tolerance mechanisms.The results show that the growth rate and biomass of the highly resistant strain A041(paraphoma chrysanthemicola)were significantly higher than those of other strains under different concentrations of Cd stress.The main resistance physiological indicators of A041 were significantly improved and different from the control(P<0.05),and with the increase of Cd concentration,the activities of catalase(CAT),peroxidase(POD)and superoxide dismutase(SOD)reached the maximum at Cd concentrations of 3 mmol/L,5 mmol/L,and 6 mmol/L,respectively,which were 215%,92%and 122%higher than those of the control group.The contents of malondial-dehyde(MDA)and soluble protein were the highest at Cd concentration of 7 mmol/L,with increases of 193%and 47%,respectively,compared with the control.Glutathione(GSH)content increased first and then decreased,reaching a peak at Cd concentration of 2 mmol/L,with an increase of 42%com-pared with the control;melanin content in A041 mycelium showed a trend of increasing first and then decreasing with the increase of Cd stress,with the highest content at Cd concentration of 2 mmol/L,with an increase of 34%compared with the control.Different concentrations of Cd stress could change the mycelial morphology of strain A041.As Cd concentration increased,the mycelial diam-eter decreased and mycelia became increasingly sparse.It is concluded that strain A041(P.chrysan-themicola)has a strong Cd tolerance.
