为探究松毛虫赤眼蜂卵黄原蛋白受体(Vitellogenin receptor,VgR)基因在其生殖调控中的作用,克隆并分析松毛虫赤眼蜂 VgR(TdVgR)基因序列,利用实时荧光定量 PCR 技术对该基因时空表达进行测定,并采用 RNA 干扰技术研究 VgR 对松毛虫赤眼蜂卵巢发育及产卵量的影响.结果表明:TdVgR cDNA序列全长5 998 bp,开放阅读框长 5 097 bp,编码氨基酸1 698个,预测编码蛋白大小 187.5 kDa.TdVgR 在松毛虫赤眼蜂整个发育阶段均表达,羽化 12 h 时表达量达到峰值.TdVgR基因在松毛虫赤眼蜂雌成虫卵巢中特异性高表达.沉默 TdVgR 后,松毛虫赤眼蜂表现出卵巢结构松散,卵子排列不规则,并出现畸形卵子,同时单蜂怀卵量及产卵量均显著降低,该结果为进一步探究松毛虫赤眼蜂的生殖调控机理提供理论依据.

To explore the role of vitellogenin receptor(VgR)gene in the reproductive regulation of Trichogramma dendrolimi,the VgR gene sequence of T.dendrolimi was cloned and analyzed,and the spatio-temporal expression of TdVgR was determined by real-time PCR.The effects of VgR on ovar-ian development and fecundity of T.dendrolimi were studied by RNA interference.The results showed that the full-length of TdVgR cDNA was 5 998 bp,with an open reading frame of 5 097 bp en-coding 1 698 amino acids,and the predicted molecular weight of the encoded protein was 187.5 kDa.TdVgR was expressed in the whole developmental stages of T.dendrolimi,and the expression level reached a peak at 12 hours after eclosion.TdVgR gene was specifically highly expressed in the ovary of female adults of T.dendrolimi.After silencing TdVgR,T.dendrolimi showed loose ovarian structure,irregular egg arrangement and deformed eggs.At the same time,the single fecundity and the number of host egg parasitiezed per female also decreased significantly.The results of this study provide ba-sic theoretical data for further exploring the reproductive regulation mechanism of T.dendrolimi.
