TCP是一类含特有bHLH结构的转录因子家族,参与调节植物细胞的发育和非生物胁迫.在干旱胁迫下利用荧光定量表达分析方法从TCP转录因子家族中筛选出显著下调的目的基因ZmTCP14,结合生物信息学对ZmTCP14基因进行生物结构和功能的预测分析,然后克隆ZmTCP14基因并构建植物过表达载体.结果表明:ZmTCP14与高粱(Sorghum bicolor)、水稻(Oryza sativa)、谷子(Setaria italica)相关性较高,有20个Mo-tif,具有高度保守结构域,同源蛋白比对与进化树分析结果一致.该蛋白是定位在细胞核上的一类亲水性蛋白,并且含有多个磷酸化位点,但不含脂蛋白信号肽和跨膜结构,启动子元件序列分析发现ZmTCP14基因与逆境胁迫和光应答之间存在密切联系.试验成功克隆出2 267 bp的单一目的条带,并构建了植物过表达载体p CAMBIA3301-ZmTCP14-Bar,为后续探索植物干旱相关基因的调控机制奠定基础.

TCP is a family of transcription factors unique to plants containing bHLH structures,in-volved in the regulation of development and abiotic stress in plant cells.In this study,a significantly down-regulated target gene ZmTCP14 was screened from TCP transcription factor family by fluores-cence quantitative expression analysis under drought stress.Combined with bioinformatics,the bio-logical structure and function prediction analysis of ZmTCP14 gene were carried out,and finally,the ZmTCP14 gene was cloned and a plant overexpression vector was constructed.The results showed that ZmTCP14 was highly correlated with sorghum(Sorghum bicolor),rice(Oryza sativa)and millet(Setaria italica).It contained 20 motifs with highly conserved domains,and the homologous protein alignment was consistent with the phylogenetic tree analysis results.This protein was a class of hy-drophilic proteins located in the nucleus,and contained multiple phosphorylation sites,but no lipo-protein signal peptides and transmembrane structures.Analysis of the promoter element sequences revealed a close relationship between ZmTCP14 gene and stress and light response.Finally,a single target band of 2 267 bp was successfully cloned,and the plant overexpression vector pCAMBIA3301-ZmTCP14-Bar was constructed.This study lays a foundation for the subsequent exploration of the regulatory mechanism of plant drought-related genes.
