为探究不同基因型与乌苏里貉总产仔数的关系,采用PCR-SSCP法对38只乌苏里貉的视黄醇结合蛋白4(RBP4)基因及催乳素受体(PRLR)基因进行多态性分析.结果表明:乌苏里貉RBP4基因存在1个多态性位点(g.42521676 C>G),PRLR基因存在1个多态性位点(g.150456010 G>A),均处于Hardy-Weinberg不平衡状态.乌苏里貉群体RBP4和PRLR基因的优势基因型均为AA基因型,优势等位基因均为A等位基因,其中RBP4基因多态位点的AA基因型频率为0.500,BB基因型频率为0.184,AB基因型频率为0.316,A和B等位基因频率分别为0.658和0.342,不同基因型乌苏里貉的总产仔数存在显著差异,AA基因型比BB基因型的总产仔数高4.6.PRLR基因多态位点的AA基因型频率为0.711,AB基因型频率为0.289,A和B等位基因频率分别为0.856和0.144,不同基因型乌苏里貉的总产仔数存在显著差异,AA基因型比AB基因型的总产仔数高3.6.研究结果为乌苏里貉早期选育的分子标记筛选提供理论依据.

To investigate the relationship between different genotypes and the total litter size of Ussuri raccoon dogs,the PCR-SSCP method was employed to analyze the polymorphisms of the retinol-binding protein 4(RBP4)gene and the prolactin receptor(PRLR)gene in 38 Ussuri raccoon dogs.The results showed that one polymorphic site(g.42521676 C>G)was identified in the RBP4 gene and one(g.150456010 G>A)in the PRLR gene,both of which were in a state of Hardy-Weinberg disequilibrium.The dominant genotype for both the RBP4 and PRLR genes in the Ussuri raccoon dog population was the AA genotype,with the A allele being the dominant allele.For the RBP4 gene polymorphic site,the frequencies of the AA,BB,and AB genotypes were 0.500,0.184,and 0.316,respectively,while the frequencies of the A and B alleles were 0.658 and 0.342,respectively.Significant differences in total litter size were observed among different genotypes,with the AA genotype having 4.6 more offspring than the BB genotype.For the PRLR gene polymorphic site,the frequencies of the AA and AB genotypes were 0.711 and 0.289,respectively,and the frequencies of the A and B alleles were 0.856 and 0.144,respectively.The AA genotype had 3.6 more offspring than the AB genotype.The results provide a theoretical basis for the screening of molecular markers for early breeding selection in this species.
