长江口葛氏长臂虾肠道菌群结构及其潜在功能的研究OA

为了解葛氏长臂虾(Palaemon gravieri)肠道菌群结构及其潜在功能,对2022年8月在长江口水域(31°06'00″N~31°24'00″N、122°00'00″E~122°14'30″E)采集到的葛氏长臂虾进行高通量测序,并对其肠道菌群的功能进行KEGG预测.结果表明,长江口葛氏长臂虾肠道菌群高质量序列条数在33 054~37 110条之间,通过聚类分析共得到 367条 OTU(分类操作单元),葛氏长臂虾肠道菌群门类以变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)为主;长江口葛氏长臂虾肠道菌群的优势菌属为嗜冷杆菌属(Psychrobacter)、巨 大 球 菌 属(Macrococcus)、分 枝 杆 菌 属(Mycobacterium)和 慢 生 根 瘤 菌 属(Bradyrhizobium).根据KEGG通路分析在各个水平最富集的通路为新陈代谢,其中主要以嘧啶代谢、丙酸代谢、氨基酸、嘌呤代谢为主,此外葛氏长臂虾肠道菌群还具有细胞进程、环境信息处理、遗传信息处理、疾病等功能.根据肠道菌群结构分析及KEGG潜在功能预测结果,初步推断长江口葛氏长臂虾会摄食含有动植物蛋白的食物、碳水化合物和一些有机碎屑.研究结果可为长江口葛氏长臂虾健康养殖、环境生态及野生繁育提供参考,也为高通量测序技术的应用提供参考.

To gain insights into the gut microbial community structure and its potential functions of Palaemon graviera,high-throughput sequencing technology was employed to analyze samples collected from the Yangtze River Estuary(31°06'00″N to 31°24'00″N,122°00'00″E to 122°14'30″E)in August 2022.Further,Kyoto Encyclopedia of Genes and Genomes predictions were performed to elucidate the functional roles of the gut microbiota.The results revealed that the number of high-quality sequencing reads ranged from 33 054 to 37 110 for Palaemon graviera from the Tangtze Estuary.Through clustering analysis,a total of 367 operational taxonomic units were identified.The gut microbial communities of Palaemon graviera were dominated by Proteobacteria and Firmicutes.And Psychrobacter,Macrococcus,Mycobacterium,and Bradyrhizobium emerged as the dominant genera.According to the Kyoto Encyclopedia of Genes and Genomes pathway analysis,the most enriched pathways at various levels were metabolism,with pyrimidine metabolism,propionate metabolism,amino acid metabolism,and purine metabolism being the primary ones.In addition,the intestinal flora of Palaemon graviera also possessed functions related to cellular processes,environmental information processing,genetic information processing,and disease.According to the gut microbiota structural analysis and Kyoto Encyclopedia of Genes and Genomes based functional predictions,it is tentatively inferred that the Palaemon graviera in the Yangtze River Estuary feeds on diets containing both animal and plant proteins,carbohydrates,as well as some organic detritus.This study lays a foundation for future research on healthy aquaculture,environmental ecology,and wild breeding of Palaemon graviera in the Yangtze River Estuary.Additionally,it contributes to the literature on the application and development of high-throughput sequencing technologies.
