山杨PdNAC2基因序列特征及表达分析OA
Sequence Characteristics and Expression Analysis of PdNAC2 Gene in Populus davidiana
为了分析山杨PdNAC2的表达模式,进一步获得山杨抗旱株系提供候选基因,以前期从山杨干旱转录组中筛选到的PdNAC2基因为研究对象,以干旱胁迫处理后的山杨叶片cDNA为模板,PCR扩增得到Pd-NAC2基因目的片段.对PdNAC2基因进行生物信息学分析,研究基因序列特征,并利用qRT-PCR研究PdNAC2的表达模式.结果表明,PdNAC2基因开放读码框852 bp,推测其编码284个氨基酸,经过多序列比对,看到PdNAC2保守结构域序列与其他物种同类的PdNAC2蛋白有着非常高的序列一致性,根据不同基因家族保守结构的不同发现山杨PdNAC2与拟南芥亲缘关系较近.通过qRT-PCR分析PdNAC2的表达模式,结果显示,随着胁迫时间的变化,PdNAC2基因的相对表达量存在一定的波动性,在第8天时PdNAC2基因的相对表达量呈现先上升后下降的趋势,PdNAC2基因的表达能够响应干旱处理,该基因可能与抗干旱胁迫相关.
In order to analyze the expression pattern of PdNAC2 in Populus davidiana and obtain candidate genes for drought-resistant strains,the PdNAC2 gene,which had been previously screened from the drought transcriptome of P.davidiana,was selected as the research object in this study.The cDNA isolated from P.davidiana leaves treated with drought stress was used as the template to amplify the target fragment of the PdNAC2 gene via PCR.Bioinformatics analysis was performed on the PdNAC2 gene to characterize its sequence features,and the expression pattern of PdNAC2 was investigated using qRT-PCR.The results showed that the open reading frame(ORF)of the PdNAC2 gene was 852 bp in length,and it was predicted to encode 284 amino acids.Multiple sequence alignment revealed that the conserved domain sequence of PdNAC2 shared extremely high sequence identity with NAC2 proteins from other species.Based on the differences in conserved structures among different gene families,PdNAC2 from P.davidiana was found to be closely related to that of Arabidopsis thaliana.Analysis of the expression pattern via qRT-PCR indicated that the relative expression level of the PdNAC2 gene fluctuated with the duration of stress.On the 8th day of stress,the relative expression level of the PdNAC2 gene exhibited an initial upward trend followed by a downward trend.Collectively,the expression of the PdNAC2 gene could respond to drought treatment,and this gene might be associated with drought resistance in P.davidiana.
