树莓Ri3GT基因克隆及表达模式分析OA
Gene Cloning and Expression Pattern Analysis of Ri3GT in Raspberry
为研究树莓类黄酮 3-O-葡萄糖基转移酶(Ri3GT)基因序列特征、表达规律和启动子结合元件,以采后树莓果实为实验材料,利用 PCR法克隆 Ri3GT基因及启动子序列并进行生物信息学分析,通过荧光定量 PCR法分析 Ri3GT表达模式.结果表明,Ri3GT 基因 cDNA 全长为 1 395 bp,可编码 464个氨基酸,有多个磷酸化位点,无信号肽.该蛋白属于 UDP-葡萄糖基转移酶家族成员,具有 PSPG 结构域,与草莓 3GT 亲缘关系最近,与矢车菊素主要通过传统氢键、Pi-Cation 作用力等 5 种作用力结合.Ri3GT表达水平在采后树莓贮藏第2d 时显著升高,该变化趋势与花青素含量变化呈正相关.树莓 Ri3GT启动子上存在光和茉莉酸甲酯响应元件.研究结果为解析 Ri3GT基因在采后树莓果实花青素合成中的作用提供一定参考.
To investigate the sequence characteristics,expression pattern,and promoter cis-elements of the raspberry flavonoid 3-O-glucosyltransferase gene(Ri3GT),postharvest raspberry fruits were used as experimental materials.The Ri3GT gene and its promoter sequence were cloned by PCR and subjected to bioinformatic anaylsis,and the expression pattern of Ri3GT was detected by quantitative real-time PCR.The results showed that the full-length cDNA of Ri3GT was 1 395 bp,encoding 464 amino acids,with multiple phosphorylation sites and no signal peptide.The protein belonged to the UDP glucosyltransferase family and contained a PSPG domain.It had the closest genetic relationship with strawberry 3GT,and bound to cyanidin mainly through five types of interactions,including conventional hydrogen bonds and Pi-Cation interactions.The expression level of Ri3GT increased significantly on day 2 of postharvest storage,and this trend was positively correlated with anthocyanin content.Light and methyl jasmonate response elements were identified in the Ri3GT promoter.These results provide a reference for elucidating the role of the Ri3GT gene in anthocyanin biosynthesis in postharvest raspberry fruits.
