几丁质合成酶基因CHSⅢ过表达及其对里氏木霉纤维素酶分泌的影响OA
Overexpression of chitin synthase CHSⅢ gene and its effect on cellulase secretion of Trichoderma reesei
[目的]探究几丁质合成酶基因CHSⅢ对里氏木霉QM6a细胞壁中几丁质质量分数及纤维素降解酶外泌的影响,为纤维素酶的工业生产菌株的构建提供理论基础.[方法]从里氏木霉中扩增几丁质合成酶基因CHSⅢ,构建CHSⅢ基因的过表达质粒,将其转化到QM6a原生质体中,筛选获得CHSⅢ基因过表达菌株,分析CHSⅢ基因对生物量、菌丝分支、细胞壁中几丁质质量分数及纤维素降解酶分泌的影响.[结果]在几丁质合成酶过表达菌株里氏木霉OE-3和OE-7中,CHSⅢ基因相对表达量明显提升;培养3 d时,OE-3和OE-7的生物量分别比里氏木霉QM6a增加20.63%和14.02%,过表达菌株的菌丝具有明显的顶端伸长现象,菌丝分支数明显少于出发菌株QM6a;OE-3和OE-7细胞壁中的几丁质质量分数比QM6a分别提高18.88%和13.06%;与之相应,整个发酵过程中OE-3和OE-7的胞外蛋白质量浓度低于QM6a,胞内蛋白质量浓度高于QM6a;OE-3和OE-7外泌的纤维素降解酶总活力比QM6a分别降低30.9%和56.5%.[结论]里氏木霉几丁质合成酶基因CHSⅢ增加里氏木霉细胞壁中几丁质质量分数,阻碍纤维素降解酶的分泌.
[Objective]This study aimed to investigate the effect of the chitin synthase gene(CHSⅢ)on the chitin mass fraction in the cell wall of Trichoderma reesei QM6a and the excretion of cellulose-degrading enzymes,so as to provide a theoretical basis for the construction of industrial cellulase-producing strains.[Method]The CHSⅢ gene was amplified from T.reesei,and an overexpression plas-mid of CHSⅢ was constructed and transformed into QM6a protoplasts.The CHSⅢ gene overexpression strains were obtained by screening,and the effects of CHSⅢ gene on biomass,hyphal branching,chitin mass fraction in the cell wall and cellulose-degrading enzyme secretion were analyzed.[Result]The relative expression level of CHSⅢ gene was significantly increased in the chitin synthase overex-pression strains T.reesei OE-3 and OE-7.After 3 days of culture,the biomass of OE-3 and OE-7 increased by 20.63%and 14.02%compared with T.reesei QM6a,respectively.The hyphae of the overexpression strains showed obvious apical elongation,and the number of hyphal branches was sig-nificantly less than that of the original strain QM6a.The chitin mass fraction in the cell wall of OE-3 and OE-7 was 18.88%and 13.06%higher than that of QM6a,respectively.Correspondingly,the extracellular protein mass concentration of OE-3 and OE-7 was lower than that of QM6a during the whole fermentation process,while the intracellular protein mass concentration was higher than that of QM6a.The total activity of excreted cellulose-degrading enzymes of OE-3 and OE-7 decreased by 30.9%and 56.5%compared with QM6a,respectively.[Conclusion]The chitin synthase gene CHSⅢin T.reesei increases the chitin mass fraction in the cell wall of T.reesei and hinders the secretion of cellulose-degrading enzymes.
