[目的]探究葡萄糖调控因子(包括胰岛素、葡萄糖处理及营养限制)对肉鸡血糖和葡萄糖转运蛋白4(glu-cose transporter 4,GLUT4)表达的影响,为进一步揭示家禽葡萄糖稳态调控机制提供理论依据,并为改善肉鸡生长性能和肌肉品质提供潜在靶点.[方法]在活体水平上,对爱拔益加(Arbor Acres,AA)肉鸡进行胰岛素(5 IU·kg-1)或葡萄糖(2 g·kg-1)腹腔注射、不同强度限饲(15%能量、15%蛋白、30%能量)和24 h禁食处理,测定血糖浓度,并通过实时反转录聚合酶链反应测定AA肉鸡不同器官或组织GLUT4基因表达.在细胞水平上,对鸡原代骨骼肌成肌细胞用不同浓度胰岛素(0、0.02、0.10、0.50、2.50 µmol·L-1)、不同质量浓度葡萄糖(0、0.5、1.0、1.5、2.5 g·L-1)分别处理120 min、12 h,以及进行葡萄糖或血清饥饿处理18 h,测定GLUT4基因表达.[结果]1)活体水平:胰岛素注射120 min后,AA肉鸡血糖极显著降低,胸肌GLUT4基因表达显著升高,腿肌极显著升高,240 min后均恢复至基础水平,肝脏无显著变化;葡萄糖注射10 min后,血糖极显著升高,胸肌和肝脏GLUT4基因表达极显著降低,60 min后均恢复,腿肌无显著变化;30%能量限饲、15%能量限饲及15%蛋白限饲对胸肌GLUT4基因表达无显著影响,24 h禁食则显著降低其表达.2)细胞水平:鸡原代成肌细胞中GLUT4基因表达呈胰岛素、葡萄糖质量浓度依赖性,0.10 µmol·L-1胰岛素处理组对GLUT4基因表达的提升效果最显著,0.5~1.0 g·L-1葡萄糖处理组基因表达到达峰值且显著高于2.5 g·L-1高糖组;葡萄糖饥饿对GLUT4基因表达无显著影响,血清饥饿则极显著上调其表达.3)胸肌GLUT4基因表达与血糖浓度呈极显著负相关,腿肌和肝脏GLUT4基因表达与血糖浓度无显著相关性.[结论]肉鸡GLUT4基因表达受胰岛素、葡萄糖及营养状态(尤其是禁食)调控,并与血糖稳态密切相关.

[Objective]This study investigated the effects of glucose regulatory factors(insulin,glucose treatment,and nutrient restriction)on blood glucose concentration and glucose transporter 4(GLUT4)expression in broilers,aiming to provide a theoretical basis for further elucidating the regulatory mecha-nisms of glucose homeostasis in poultry,and to offer potential targets for improving broiler growth perfor-mance and muscle quality.[Method]At the in vivo level,Arbor Acres(AA)broiler chickens were administered with intraperitoneal injections of insulin(5 IU·kg-1)or glucose(2 g·kg-1),subjected to different levels of feed restriction(15%energy restriction,15%protein restriction,30%energy restric-tion),or fasted for 24 hours.Blood glucose was measured at specific time points,and GLUT4 gene expression in various tissues was determined using qRT-PCR.At the cellular level,primary chicken skeletal myoblasts were treated with different concentrations of insulin(0,0.02,0.10,0.50,2.50 µmol·L-1)for 120 min or glucose(0,0.5,1.0,1.5,2.5 g·L-1)for 12 h.In a separate experiment,cells were subjected to glucose or serum starvation for 18 h,after which GLUT4 gene expression was mea-sured.[Result]1)In vivo:At 120 min post-insulin injection,blood glucose was significantly decreased,while GLUT4 gene expression was significantly increased in pectoral muscle and an extremely significant increase in the leg muscle,both returning to basal levels by 240 min;No significant changes were observed in the liver.At 10 min post-glucose injection,blood glucose was significantly elevated,and GLUT4 gene expression in pectoral muscle and liver was extremely significantly decreased,recove-ring by 60 min,with no significant change in leg muscle.Feed restriction(30%energy,15%energy,or 15%protein)had no significant effect on GLUT4 gene expression in pectoral muscle,whereas 24 h fas-ting significantly reduced its expression.2)In vitro:In chicken primary myoblasts,GLUT4 gene expre-ssion exhibited a concentration-dependent response to both insulin and glucose.The most significant increase was observed with 0.10 µmol·L-1 insulin treatment.Expression peaked in the 0.5-1.0 g·L-1 glucose treatment groups and was significantly higher than in the 2.5 g·L-1 high-glucose group.Glucose starvation had no significant effect on its expression,whereas serum starvation significantly upregulated its expression.3)A significant negative correlation was found between GLUT4 gene expression in pecto-ral muscle and blood glucose concentration,while no such correlation was observed for leg muscle or liver.[Conclusion]Chicken GLUT4 gene expression is regulated by insulin,glucose,and nutritional status(notably fasting)and is closely associated with blood glucose homeostasis.
