[目的]鉴定毛白杨RBOH(respiratory burst oxidase homologue)家族成员,探究其在毛白杨生长发育中的功能和作用,并为毛白杨的抗逆改良提供信息基础.[方法]通过对毛白杨全基因组数据进行系统分析,鉴定到19个RBOH家族成员,并对其进化关系、基因结构、保守基序与结构域、启动子顺式作用元件和时空表达模式等进行研究.[结果]毛白杨RBOH家族成员分为5个亚族,分布在11条染色体上.进化分析显示,毛白杨RBOH家族成员与拟南芥和水稻的RBOH成员具有较高的同源性.顺式作用元件分析表明,毛白杨的启动子区域包含多种响应光、激素和胁迫信号的元件,表明其可能参与多种生物学过程.时空表达模式显示,PtoRboh4A在毛白杨根中优先高丰度表达,PtoRboh11D的表达随茎的木质化程度逐渐升高,PtoRboh2D和PtoRboh3D在成熟叶中优先高表达;在茎中,PtoRboh4A在韧皮部优先表达,PtoRboh1A、PtoRboh1D和PtoRboh5A在木质部中优先表达.[结论]系统解析了毛白杨RBOH家族成员的特征和表达模式,其家族成员序列和进化相对保守,功能具有多样性.其中PtoRboh2D、PtoRboh3D、PtoRboh4A和PtoRboh11D等基因的表达模式暗示其可能具有重要的生物学功能.

[Objective]This study was conducted to identify members of the RBOH(respiratory burst oxidase homologue)family in Populus tomentosa and investigate their functions and roles in the growth and development of this species,thereby providing an informational foundation for stress resistance improvement in Populus tomentosa.[Method]The whole genome data of Populus tomentosa were sys-tematically analyzed,19 members of the RBOH family were identified,and their evolutionary relation-ships,gene structures,conserved motifs and domains,promoter cis-acting elements,and spatiotempo-ral expression levels were studied.[Results]The RBOH family members of Populus tomentosa are divided into 5 subfamilies,distributed on 11 chromosomes.Evolutionary analysis shows that RBOH members of Populus tomentosa have high homology with those of Arabidopsis and rice.The analysis of cis-acting elements shows that the promoter region of PtoRbohs in Populus tomentosa contains multiple elements that respond to light,hormones,and stress signals,indicating their possible involvement in various biological processes.Temporal and spatial expression analysis showed that PtoRboh4A was highly expressed in the roots of Populus tomentosa,while PtoRboh11D expression gradually increased with the degree of stem lignification.PtoRboh2D and PtoRboh3D were preferentially expressed in mature leaves.In the stem,PtoRboh4A was preferentially expressed in the phloem;PtoRboh1A,PtoR-boh1D and PtoRboh5A were preferentially expressed in the xylem.[Conclusion]The characteristics and expression patterns of the RBOH family members in Populus tomentosa were systematically ana-lyzed.The sequence and evolution of the RBOH family members in Populus tomentosa were relatively conservative,and they exhibited functional diversity.The high expression patterns of genes such as PtoRboh2D,PtoRboh3D,PtoRboh4A,and PtoRboh11D suggest that they may have important biologi-cal functions.
