目的:评价小檗碱(BBR)与表没食子儿茶素没食子酸酯(EGCG)自组装纳米复合物 BE 对小鼠急性溃疡性结肠炎(UC)的治疗效果.方法:采用纳米沉淀法制备 BE,观察 BBR、EGCG 及 BE 在水中的溶液状态.30 只6周龄雄性 C57BL/6J 小鼠随机分为6 组,每组5 只.空白组饮用普通饮用水,并每日灌胃 100 μL 蒸馏水;模型组、BBR 组、EGCG 组、B+E 组、BE 组饮用葡聚糖硫酸钠溶液(诱导 UC),同时每天分别灌胃蒸馏水、40 mg/kg BBR、60 mg/kg EGCG、50 mg/kg BBR 与 EGCG 物理混合溶液和50 mg/kg BE 溶液100 μL;共持续7 d.第9 天处死小鼠,分离结肠,评估疾病活动指数(DAI)、结肠长度、脾脏指数,采用 HE 和天狼星红染色行组织病理学观察;采用 RT-qPCR 检测结肠组织中 IL-6、TNF-α、IL-1β mRNA 表达水平.结果:BBR 在水中呈黄色悬浮状态,EGCG 水溶液无色透明,BE 水溶液为均匀的浅黄色澄清液体,且无明显沉淀或分层.实验期间空白组 DAI 为0;其余5 组小鼠体重下降,DAI 评分增加,BE 组体重和 DAI 变化幅度小于其他4 组(P<0.05).与空白组比较,模型组结肠长度缩短;纤维化面积百分比,IL-6、TNF-α、IL-1β mRNA 表达水平及脾脏指数均升高(P<0.05).与模型组比较,BBR 组、EGCG组、B+E 组及 BE 组上述指标及病理学综合评分均有下降趋势,且 BE 组变化最显著(P<0.05).结论:BE 水溶性良好,可有效缓解 UC,效果优于 BBR、EGCG 单药及两者的物理混合.

Aim:To evaluate the therapeutic effect of self-assembled nanocomposite(BE)of berberine(BBR)and epi-gallocatechin gallate(EGCG)on acute ulcerative colitis(UC)in mice.Methods:BE was prepared by nanoprecipitation,and the states of dissolution in water of BBR,EGCG and BE were observed.Thirty 6-week-old male C57BL/6J mice were randomly divided into 6 groups(5 in each group).Mice in the blank group were given ordinary drinking water and intragas-trically administered 100 μL distilled water daily.Mice in the model group,BBR group,EGCG group,B+E group and BE group were given dextran sulfate sodium solution,and were intragastrically administered 100 μL distilled water,40 mg/kg BBR,60 mg/kg EGCG,50 mg/kg physical mixture of BBR and EGCG,and 50 mg/kg BE solution daily,respectively,for 7 consecutive days.Mice were sacrificed on day 9,and colons were isolated.Disease activity index(DAI),colon length and spleen index were evaluated.Histopathological analysis was performed by HE and picrosirius red staining.The mRNA ex-pression levels of IL-6,TNF-α and IL-1β in colon tissues were detected by RT-qPCR.Results:BBR appeared as a yellow suspension in water,EGCG solution was colorless and transparent,and BE solution was a uniform pale yellow clear liquid without obvious precipitation or stratification.During the experiment,DAI of the blank group was 0.Mice in the other 5 groups showed body weight loss and increased DAI scores,and the changes in body weight and DAI in the BE group were smaller than those in the other 4 groups(P<0.05).Compared with the blank group,the model group showed shortened co-lon length,elevated percentage of fibrotic area,mRNA expression levels of IL-6,TNF-α and IL-1β,as well as spleen index(P<0.05).Compared with the model group,the above indicators and comprehensive pathological score in the BBR,EGCG,B+E and BE groups all showed a downward trend,and the changes were most significant in the BE group(P<0.05).Conclusion:BE has good water solubility,can effectively alleviate UC,and its efficacy is superior to BBR,EGCG a-lone and their physical combination.
