Relation between the expression of ZEB1 and PD-L1 in gastric cancer tissues and cells

目的:探讨胃癌组织及细胞中锌指 E 盒结合同源盒 1(ZEB1)和程序性死亡配体1(PD-L1)表达的关系.方法:采用 RNAscope 技术检测94 例胃癌组织芯片(含86 例配对胃癌与癌旁正常组织)中 ZEB1 mRNA 表达水平,采用免疫组化法检测 PD-L1 蛋白的表达.将胃癌细胞系 BGC-823 及 AGS 分为对照组、干扰组,使用 Lipofectamine 3000 分别转染无义序列、ZEB1-siRNA,48 h 后分别采用 RT-qPCR、Western blot 及免疫荧光法检测 ZEB1 和 PD-L1 mRNA、蛋白及细胞表面 PD-L1 的表达水平.结果:ZEB1 mRNA 在胃癌细胞及间质中均有表达,PD-L1 蛋白主要定位于胃癌间质,少量表达于胃癌细胞表面;胃癌组织中 ZEB1 mRNA 和 PD-L1 蛋白阳性表达率均高于癌旁正常组织(ZEB1:96.5%vs 86.0%,P=0.004;PD-L1:88.4%vs 29.1%,P<0.001),且两者表达正相关(rS=0.260,P=0.011).干扰组和对照组 BGC-823、AGS 细胞中 ZEB1 和 PD-L1 的 mRNA、蛋白表达水平,细胞表面 PD-L1 表达均低于对照组(P<0.05).结论:胃癌组织中 ZEB1、PD-L1 高表达,胃癌细胞中 ZEB1 对 PD-L1 具有正向调控作用.

Aim:To investigate the relation between zinc finger E-box binding homology box 1(ZEB1)and pro-grammed cell death-ligand 1(PD-L1)expression in gastric cancer tissues and cells.Methods:The expression levels of ZEB1 mRNA in 94 gastric cancer tissue microarrays(including 86 paired gastric cancer tissues and adjacent normal tissues)were detected using RNAscope technology,and PD-L1 protein expression was assessed by immunohistochemistry.Gastric cancer cell lines BGC-823 and AGS were divided into control group and interference group,and transfected with nonsense sequence or ZEB1-siRNA using Lipofectamine 3000.After 48 hours,the expression levels of ZEB1 and PD-L1 mRNA,pro-tein,and cell surface PD-L1 were detected by RT-qPCR,Western blot,and immunofluorescence,respectively.Results:ZEB1 was expressed in both gastric cancer cells and stroma,while PD-L1 was primarily localized in the gastric cancer stroma with minimal expression on the surface of gastric cancer cells.The positive expression rates of ZEB1 mRNA and PD-L1 protein in gastric cancer tissues were significantly higher than those in adjacent normal tissues(ZEB1:96.5%vs 86.0%,P=0.004;PD-L1:88.4%vs 29.1%,P<0.001),and showed a positive relation(rS=0.260,P=0.011)between them.The mRNA and protein expression levels of ZEB1 and PD-L1,as well as the cell surface PD-L1 expression of BGC-823 and AGS cells in the intervention group were significantly lower than those in the control group(P<0.05).Conclusion:ZEB1 and PD-L1 are highly expressed in gastric cancer tissues,and ZEB1 exerts a positive regulatory effect on PD-L1 in gastric cancer cells.
