目的:本研究旨在探讨电针干预对急性脑梗死大鼠 NLRP3/caspase-1 信号通路和血-脑屏障相关因子表达的影响,以阐明其潜在的神经保护机制.方法:采用大脑中动脉闭塞法 MCAO 大鼠模型,随机分为空白对照组、模型组(MCAO)、假电针干预组(MCAO+sham EA)和电针干预组(MCAO+EA).通过 TTC 染色评估脑梗死体积;采用 H&E 及尼氏染色观察组织病理学改变与神经元凋亡,利用免疫荧光染色检测神经元凋亡、小胶质细胞及星形胶质细胞活化情况;采用免疫组化法分析神经修复相关因子表达;并运用 qRT-PCR 及 WB 检测脑组织中 NLRP3/caspase-1 信号通路相关因子及血-脑屏障紧密连接蛋白 ZO-1、Claudin-5 的表达水平.结果:电针干预能极显著减小 MCAO 大鼠脑梗死体积(P<0.01),减轻神经炎症反应,并抑制神经元凋亡;同时,电针可以显著下调 NLRP3、caspase-1 等炎症通路因子的表达(P<0.05),抑制小胶质细胞与星形胶质细胞的过度活化.此外,电针可上调紧密连接蛋白ZO-1 与 Claudin-5 的表达(P<0.05),减轻血-脑屏障损伤,并促进脑源性神经营养因子与碱性成纤维细胞生长因子等神经修复相关因子的表达.结论:电针可能通过抑制神经胶质细胞活化、调控 NLRP3/caspase-1 信号通路及增强血-脑屏障完整性,发挥抗炎与神经保护作用,从而缓解 MCAO 诱导的脑组织损伤.

Objective:To investigate the effects of electroacupuncture(EA)intervention on the NL-RP3/caspase-1 signaling pathway and the expression of blood-brain barrier(BBB)-related factors in rats with acute cerebral infarction,so as to elucidate its potential neuroprotective mechanisms.Methods:A rat model of middle cerebral artery occlusion(MCAO)was established and the rats were randomly divided into a blank control group,model group(MCAO),sham electroacupuncture intervention group(MCAO+sham EA),and electroacupuncture intervention group(MCAO+EA).Cerebral infarction volume was assessed by TTC staining;histopathological changes and neuronal apoptosis were observed by H&E and Nissl staining;neuronal apoptosis,microglial activation,and astrocyte activation were detected by immunofluorescence stai-ning;the expression of neurorepair-related factors was analyzed by immunohistochemistry;and the expression of NLRP3/caspase-1 signaling pathway-related factors as well as BBB tight junction proteins ZO-1 and Clau-din-5 in brain tissue were measured by qRT-PCR and Western blot(WB).Results:EA intervention signifi-cantly reduced the cerebral infarction volume in MCAO rats(P<0.01),alleviated neuroinflammation,and in-hibited neuronal apoptosis.Meanwhile,EA significantly downregulated the expression levels of inflammatory pathway factors such as NLRP3 and caspase-1(P<0.05),and inhibited the overactivation of microglia and astrocytes.Furthermore,EA upregulated the expression levels of tight junction proteins ZO-1 and Claudin-5(P<0.05),mitigated BBB damage,and promoted the expression of neurorepair-related factors such as brain-derived neurotrophic factor(BDNF)and basic fibroblast growth factor(bFGF).Conclusion:Electroacu-puncture may exert anti-inflammatory and neuroprotective effects,thereby alleviating MCAO-induced brain tissue damage,potentially by inhibiting neuroglial cell activation,regulating the NLRP3/caspase-1 signaling pathway,and enhancing blood-brain barrier integrity.
