为阐明猪流行性腹泻病毒(Porcine epidemic diarrhea virus,PEDV)遗传变异特征,本研究对PEDV阳性样品进行病毒样品检测与分离,通过RT-PCR、间接免疫荧光及透射电镜等试验进行鉴定,并结合二代测序技术完成全基因组测序及遗传进化分析.结果显示,本研究成功得到1株PEDV分离株,命名为PEDV CH/HN/2024株.同源性比对结果表明,该分离株全基因组序列与ZQ2022株展现出极高相似性;系统发育进化树分析显示,CH/HN/2024株与ZQ2022株属于同一分支,为G2a亚型.S蛋白关键抗原表位区域进行分析显示,与经典毒株CV777相比,该毒株的S蛋白在COE区与SS6区分别展现出10个及2个氨基酸序列差异.本研究旨在为掌握当前我国PEDV流行特征、深入解析病毒遗传进化规律提供数据支撑,同时为后续PED疫苗研发提供科学依据,进而助力养猪业PED综合防控体系的完善与优化.

To clarify the genetic variation characteristics of Porcine epidemic diarrhea virus(PEDV),this study conducted detection and isolation of viruses from PEDV-positive samples,followed by identification via experiments including RT-PCR,indirect immunofluorescence assay(IFA)and transmission electron microscopy(TEM).Meanwhile,whole-genome sequencing and genetic evolution analysis were performed based on next-generation sequencing(NGS)technology.The results showed that one PEDV isolate was successfully obtained in this study,designated as PEDV CH/HN/2024 strain.Homology alignment analysis revealed that the complete of this isolate shared extremely high similarity with those of the ZQ2022 strain.Phylogenetic tree analysis indicated that the CH/HN/2024 strain clustered in the same branch as the ZQ2022 strain,belonging to the G2a subtype.Analysis of key antigenic epitope regions of the S protein demonstrated that,the S protein of this isolate exhibited 10 and 2 amino acid sequence variations in the COE and SS6 regions compared with the classic CV777 strain,respectively.This study intended to lay a data foundation for clarifying the current epidemic characteristics of PEDV and deeply deciphering the genetic evolution rules of the virus,and provide a scientific basis for subsequent PED vaccine research and development,thereby facilitating the improvement and optimization of the comprehensive prevention and control system for PED in the swine industry.
