奶牛子宫源海氏肠球菌的分离鉴定及安全性研究OA
Isolation,identification and safety evaluation of Enterococcus hirae from dairy cow uterus
本研究旨在从健康高产成年奶牛的子宫中分离并筛选优良菌株,为研发防治细菌性奶牛子宫内膜炎的微生物制剂提供潜在的候选菌株.通过16SrDNA测序对分离纯化的菌株进行菌种鉴定,系统评价分离菌株的生长特性、产酸能力、黏附性、自凝集率、表面疏水性及抑菌活性;通过抗生素敏感性试验、CCK-8细胞毒性试验和小鼠体内安全性试验评估菌株安全性,并对菌株主要抗菌物质进行初步分析.结果显示,本研究成功分离出2株海氏肠球菌,命名为A8和E5.2株菌均具有良好的产酸能力与黏附能力,能显著抑制金黄色葡萄球菌和大肠杆菌的生长,主要抑菌物质均包括过氧化氢、细菌素以及有机酸,对环丙沙星、四环素、红霉素、利福平、氨苄西林、氯霉素、青霉素等10种常用抗生素表现出较强的敏感性,对奶牛子宫内膜上皮细胞无毒性,小鼠子宫灌注试验中未见损伤.综上所述,海氏肠球菌A8和E5具有良好的益生特性与生物安全性,具备作为微生态制剂用于奶牛子宫内膜炎临床防治的潜力.
This study aimed to isolate and screen excellent strains from the uterine of healthy,high-yield adult dairy cows,thus providing potential candidate strains for the development of microbial preparations against bacterial endometritis in dairy cows.The isolated and purified strains were identified via 16S rDNA sequencing.Systematic evaluations were conducted on the isolates regarding their growth characteristics,acid-producing capacity,adhesion ability,autoaggregation rate,surface hydrophobicity,and antibacterial activity.The safety of the strains was assessed through antibiotic susceptibility test,CCK-8 cytotoxicity assay,and in vivo safety test in mice.And a preliminary analysis was performed on the main antibacterial substances produced by the strains.The results demonstrated that two strains of Enterococcus hirae were successfully isolated in this study,which were designated as A8 and E5,respectively.Both strains exhibited favorable acid-producing and adhesion capabilities,which significantly inhibited the growth of Staphylococcus aureus and Escherichia coli.The main antibacterial substances produced by the two strains included hydrogen peroxide,bacteriocins,and organic acids.Additionally,both strains showed high sensitivity to nine commonly used antibiotics,including ciprofloxacin,tetracycline,erythromycin,rifampicin,ampicillin,chloramphenicol,and penicillin.They exerted no cytotoxic effects on bovine endometrial epithelial cells,and no uterine damage was observed in the mouse uterine perfusion test.In conclusion,Enterococcus hirae A8 and E5 possess excellent probiotic properties and biosafety,thus holding great potential for being developed as probiotic preparations in the clinical prevention and treatment of bovine endometritis.
