白花泡桐光信号调控基因PfHY5的克隆与功能分析OA
Cloning and functional analysis of PfHY5 gene in Paulownia fortunei
为了初步探究白花泡桐(Paulownia fortunei)转录因子HY5的功能和表达调控机制,基于白花泡桐基因组数据库,克隆得到了PfHY5基因及其启动子序列并对其进行了生物信息学及表达分析.结果表明,PfHY5基因长度为474 bp编码157个氨基酸.PfHY5是一个结构保守的酸性蛋白,定位于细胞核中,二级结构主要由α-螺旋组成.PfHY5基因在白花泡桐茎和根中高表达,可响应光照和泡桐丛枝病.同时,也验证了 PfHY5基因上游1 993 bp的启动子序列具有转录激活活性,预测到该区域包含响应光、胁迫和激素的顺式元件,表明PfHY5基因表达受到多种因素的调控.这些结果为明晰白花泡桐PfHY5基因的功能和光信号调控途径奠定了基础,也为其他木本植物光响应机制的研究提供了参考.
To preliminarily explore the function and transcriptional regulation mechanism of a transcription factor HY5 in Paulownia fortunei,we cloned the coding and promotor sequence of PfHY5 referring to the P.fortunei genome database,followed by bioinformatics and expression analysis.The results showed that the PfHY5 gene was 474 bp in length and encoded 157 amino acids.PfHY5 is a structurally conserved acidic protein located in the nucleus,with its secondary structure mainly composed of α-helices.PfHY5 gene was highly expressed in the stem and root of P.fortunei,and responded to light and Paulownia witches' broom disease.Further,we verified the transcriptional activation activity of its 1993-bp promoter sequence.It was predicted that this region contained cis-elements responsive to light,stress and hormones,indicating that the expression of PfHY5 gene was regulated by multiple factors.These results lay the foundation for clarifying the function of the PfHY5 gene and the light signal transduction pathway in P.fortunei,and also provide a reference for the study of light response mechanism in other woody plants.
