复方扶芳藤合剂对黑色素瘤小鼠脾脏CD4+T细胞的调节作用OA
The regulatory effect of Compound Fufangteng Mixture on CD4+T cells in the spleen of melanoma mice
目的:探究复方扶芳藤合剂(compound fufangteng mixture,CFM)对黑色素瘤小鼠脾脏CD4+T细胞的调节作用.方法:将50只雌性C57BL/6小鼠随机分为5组:空白对照(Blank)组、肿瘤模型(B16)组、高剂量复方扶芳藤合剂(CFM-H)组、中剂量复方扶芳藤合剂(CFM-M)组和低剂量复方扶芳藤合剂(CFM-L)组,预先灌胃纯水或不同剂量CFM 21 d,再皮下注射B16细胞构建黑色素瘤模型.2周后终止实验,取小鼠脾脏,用流式细胞术检测T细胞比例及其活化、增殖、凋亡、抑制、转录因子表达和细胞因子分泌情况.结果:CFM对小鼠体重及胸腺指数无明显影响,中剂量CFM可提高脾脏指数;中剂量CFM降低了CD4+T细胞比例,提高了CD8+T细胞比例,对总T细胞比例无明显影响;中剂量CFM能促进Th1、Th2细胞数量增加;中剂量CFM能促进T细胞CD69和CD38的表达;高剂量CFM能促进T细胞CD95表达,中、低剂量 CFM 能促进 T细胞 PD-1表达;高剂量 CFM 促进 IL-4分泌,中剂量 CFM 促进 IL-2、IFN-γ、IL-4和IL-10分泌,低剂量CFM促进TNF-α、IL-4分泌;高、中剂量CFM能增加NF-κB表达,减少T-bet和GATA-3表达,低剂量CFM能降低T-bet表达.结论:CFM能够活化CD4+T细胞,促进Th1、Th2细胞亚群的分化.
Objective:To explore the regulatory effect of Compound Fufangfuteng Mixture(CFM)on CD4+T cells in the spleen of melanoma mice.Methods:Randomly divide 50 female C57BL/6 mice into 5 groups:blank group(Blank),tumor model group(B16),high-dose Compound Fufangfuteng Mixture group(CFM-H),medium-dose Compound Fufangfuteng Mixture group(CFM-M),and low-dose Compound Fufangfuteng Mixture group(CFM-L).Pre-treat the mice with pure water or different doses of CFM by gavage for 21 days,followed by subcutaneous injection of B16 cells to establish melanoma model.After 2 weeks,ter-minate the experiment,harvest the mouse spleens,and use flow cytometry to detect the proportion of T cells and their activation,proliferation,apoptosis,inhibition,transcription factor expression and cytokine secretion.Results:CFM had no significant effect on mouse body weight and thymus index,but the medium dose of CFM could increase the spleen index;the medium dose of CFM reduced the proportion of CD4+T cells and increased the proportion of CD8+T cells,with no significant effect on the total T cell proportion;the medium dose of CFM could promote the increase in the number of Th1 and Th2 cells;The medium dose of CFM could promote the expression of CD69 and CD38 on T cells;the high dose of CFM could promote the expression of CD95 on T cells,while the medium and low doses of CFM could promote the expression of PD-1 on T cells;The high dose of CFM could promote the secretion of IL-4,the medium dose of CFM could promote the increase of IL-2,IFN-γ,IL-4,and IL-10,and the low dose of CFM could promote the secretion of TNF-α and IL-4;both high and medium doses of CFM could increase the expres-sion of NF-κB,the medium dose of CFM could decrease the expression of T-bet and GATA-3,and the low dose of CFM could reduce the expression of T-bet.Conclusions:CFM can activate CD4+T cells and promote the differentiation of Th1 and Th2 cell subsets.
