为探究继代培养对马桑菌生长和漆酶活力的影响,以贵州特色食用菌马桑菌(Lentinula edodes)为对象,研究了继代培养对其菌丝生长特性、拮抗反应及漆酶活力的影响.结果表明,各继代菌株与F1代之间均未出现拮抗反应,仅从表观上不能判断马桑菌F15代内是否发生遗传变异.菌丝体日均生长速度随继代次数增加整体呈下降趋势,其中F1代和F7代表现最快,日均生长速度达0.95 mm/d.F1~F5代接种点附近菌落的浓密度和洁白度高于其他继代菌株,而F13代和F15代出现角变现象.F1~F9代间漆酶活力呈正向促进作用,F9代以后漆酶活性波动较大且不规律,F13代漆酶活力最大,为113.89 U/L,较F1代提高1.26倍.综合菌丝体生长与漆酶活力表现,建议在马桑菌实际菌种生产中,继代次数应控制在F5代以内,以保障菌种质量和栽培安全.

To investigate the effects of subculture on the growth and laccase activity of Lentinula edodes(a characteristic edible fungus in Guizhou Province),the mycelial growth characteristics,antagonistic reactions,and laccase activity of the fungus during subculture were studied.The results showed that no antagonistic reaction was observed between any subcul-tured strain and the F1 generation,indicating that genetic variation within the F15 generation could not be determined based solely on phenotypic observation.The daily mycelial growth rate exhibited an overall decreasing trend with increasing sub-culture generations,with the F1 and F7 generations showing the fastest growth,reaching a daily growth rate of 0.95 mm/d.The mycelial density and whiteness near the inoculation point were higher in the F1 to F5 generations compared to other subcultured strains,whereas sectoring was observed in the F13 and F15 generations.Laccase activity was positively pro-moted from F1 to F9 generations,but fluctuated more irregularly after the F9 generation.The highest laccase activity(113.89 U/L)was observed in the F13 generation,which was 1.26 times higher than that of the F1 generation.Considering both mycelial growth and laccase activity,it is recommended that the number of subculture generations should be con-trolled within F5 in the practical spawn production of Lentinula edodes to ensure spawn quality and cultivation safety.
