对甘蔗多酚提取物的抗氧化活性进行测定与评价,建立适用于甘蔗多酚的多指标联合测定方法,对各测定方法的测量不确定度来源进行系统分析与评定.采用2,2-二苯基-1-苦基肼自由基清除法(DPPH法)、2,2ʹ-联氮双(3-乙基苯并噻唑啉-6-磺酸)自由基清除法(ABTS法)和铁离子还原抗氧化能力法(FRAP法)3 种经典体外抗氧化活性测定方法,对甘蔗多酚提取物进行抗氧化活性评价,并对方法学各项指标进行系统验证.3 种测定方法线性关系良好,相关系数 R2 均大于 0.999;精密度 RSD 均小于2.0%,重复性RSD均小于 3.5%,稳定性良好;加标回收率为 97.4%~100.5%,检出限分别为 1.21、1.05 μg/mL和 5.50 μmol/L.甘蔗多酚DPPH自由基清除率为 78.32%±1.39%(k=2);ABTS自由基清除率为 85.47%±1.72%(k=2);FRAP值为(456.8±10.6)μmol/g(k=2),置信水平均约为 95%.不确定度主要来源于样品溶液制备、吸光度测量、温度控制和重复性测量等环节.建立的测定方法准确可靠,测量不确定度评定结果可为甘蔗多酚抗氧化活性的质量控制和标准化检测提供科学技术支撑.

To determine and evaluate the antioxidant activity of sugarcane polyphenol extracts,establish a multi-index combined determination method applicable to sugarcane polyphenols,and systematically analyze and evaluate the measurement uncertainty of each determination method.Three classical in vitro antioxidant activity assays—the 2,2-diphenyl-1-picrylhydrazyl(DPPH)radical scavenging method,the 2,2ʹ-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)radical scavenging method,and the ferric reducing antioxidant power(FRAP)method—were used to evaluate the antioxidant activity of sugarcane polyphenol extracts,with systematic validation of methodological parameters.All three methods showed excellent linearity,with correlation coefficients R2 greater than 0.999.Precision RSD was less than 2.0%,repeatability RSD was less than 3.5%,and stability was satisfactory.Spike recoveries ranged from 97.4%to 100.5%,and the limits of detection were 1.21,1.05 μg/mL,and 5.50 μmol/L,respectively.The DPPH radical scavenging rate was 78.32%±1.39%(k=2);ABTS radical scavenging rate was 85.47%±1.72%(k=2);and the FRAP value was(456.8±10.6)μmol/g(k=2),all at approximately 95%confidence level.The primary sources of measurement uncertainty were sample solution preparation,absorbance measurement,temperature control,and repeatability.The determination methods are accurate and reliable,and the measurement uncertainty evaluation provides technical support for quality control and standardized testing of sugarcane polyphenol antioxidant activity.
