[目的]探明草地贪夜蛾 1龄虫取食对高粱幼苗叶片和根部转录组的影响,为解析高粱幼苗应对草地贪夜蛾取食的分子机制及培育抗虫品种提供理论依据.[方法]以红粱丰 1号高粱幼苗为材料,设置对照组(CK)、草地贪夜蛾 1龄虫取食处理组(TR),处理 9 h后采集叶片和根部样品,进行转录组测序分析.[结果]叶片鉴定出 1 509个差异表达基因(1 202个上调,307个下调),根部鉴定出 2 128个差异表达基因(1 493个上调,635个下调),具有组织特异性.叶片上调基因主要参与次生代谢物合成、脂肪酸代谢、茉莉酸合成等防御相关通路,下调基因涉及DNA修复、繁殖等过程.根部上调基因富集于苯丙烷类生物合成、角质和蜡质合成及植物激素信号转导等通路,下调基因与生长、转运相关.[结论]高粱幼苗叶片通过激活防御代谢通路、优先分配防御资源应对取食胁迫,根部通过接收信号协同响应、协调资源分配参与防御.

[Objective]The effects of 1st-instar Spodoptera frugiperda feeding on leaf and root transcriptomes of sorghum seedlings were explored to provide a theoretical basis for illustrating the molecular mechanism in response of sorghum seedlings to S.frugiperda feeding and breeding insect-resistant sorghum varieties.[Method]The leaf and root samples of Hongliangfeng 1(a sorghum variety)seedlings fed by 1st-instar S.frugiperda for 9 h(TR)were analyzed for transcriptome sequencing by taking normal Hongliangfeng 1 seedlings as CK.[Result]A total of 1 509 differentially expressed genes(DEGs)of 1 202 up-regulated and 307 down-regulated DEGs were identified in leaves,and 2 128 DEGs of 1 493 up-regulated and 635 down-regulated DEGs were identified in roots,which showed that DEGs were of tissue-specificity.The up-regulated genes in leaves were primarily involved in defense-related pathways of secondary metabolite synthesis,fatty acid metabolism,jasmonic acid biosynthesis,and so on,and the down-regulated genes in leaves were associated with DNA repair and reproduction.The up-regulated genes in roots were enriched in pathways of phenylpropanoid biosynthesis,cutin and waxiness synthesis,and phytohormone signaling transduction,and the down-regulated genes were related to growth and transport process.[Conclusion]The leaves of sorghum seedlings respond to S.frugiperda feeding stress by activating defense metabolic pathways and preferentially allocating defense resource,while the roots involves in defense by signal reception,synergistic response,and resource reallocation.
