生姜组培苗后代群体的性状分析OA
Analysis on Traits of Offspring Population of Ginger Tissue Culture Seedlings
[目的]探明生姜组培变异机理,以期在苗期筛选突变植株,减少继代、移栽及后期管理成本,为生姜种质创新奠定基础.[方法]从表型性状、细胞学及分子水平分析突变植株与正常植株间差异.[结果]生姜突变植株生产种(M'3)表现为植株矮化,分枝数增多,根状茎重降低,鲜叶重无明显差异;12对SRAP引物共扩增 98条谱带,多态性比率为 77.36%,多态性信息指数在 0.646 5~0.890 0,生姜组培苗(CK)、原原种、原种、生产种群体的遗传一致度为 0.674 7~0.861 0,遗传距离为 0.149 6~0.393 5;生姜生产种正常植株(M3)与突变植株(M'3)的根尖细胞染色体数均为 22条,即 2n=2x=22,为二倍体.[结论]生姜生产种在表型特征方面发生明显变异,染色体数目无明显改变.在分子水平上,生姜组培苗后代存在较广泛的体细胞变异.
[Objective]The variation mechanism of ginger tissue culture seedlings was investigated to effectively select the mutant seedlings for reducing the cost in subculture,transplanting and later management and lay the foundation for innovation of ginger germplasm resources.[Method]The differences between mutant and normal tissue culture plants were analyzed from the angles of phenotypic traits,cytology and molecular level.[Result]The mutant plants from M'3 generation showed dwarf,more branch number and lower rhizome weight,and no significant difference in fresh leaf weight compared with the normal plant.A total of 98 bands with a polymorphism ratio of 77.36%and polymorphism information content(PIC)of 0.646 5-0.890 0 were amplified by 12 pairs of SRAP primers.The genetic identity and genetic distance of four ginger populations(tissue culture seedling,breeder's seed,original seed and production seed)were 0.674 7-0.861 0 and 0.149 6-0.393 5 separately.The chromosome number of root tip cells in normal(M3)and mutant(M'3)plants both was 22(2n=2x=22).[Conclusion]There is a significant variation in phenotypic characteristics of plants from ginger production seed and no variation in chromosome number of plants from ginger production seed.At the molecular level,there is an extensive somatic cell variation in the offspring of ginger tissue culture seedlings.
