金银花CBF/DREB1基因家族的鉴定与功能分析OA
Identification and Functional Analysis of CBF/DREB1 Gene Family in Honeysuckle
[目的]探究金银花CBF/DREB1基因家族成员的结构、组成和特征及其在不同组织和非生物胁迫下的表达模式,为提高金银花抗逆性、产量和品质提供理论依据.[方法]以拟南芥 6个CBF/DREB1基因家族成员的氨基酸序列为模板,采用生物信息学方法,在金银花蛋白质数据库中进行Blast比对,分析LjCBFs的理化性质及特征;基于NCBI数据库中的转录组数据,分析LjCBFs在不同组织中的表达模式;以金花3号为研究材料,于二白期采用不同非生物胁迫处理,采集花蕾分析LjCBFs的表达模式.[结果]在金银花中共鉴定出 16个CBF/DREB1基因家族成员,均为亲水蛋白,不含跨膜结构域和信号肽,含 1~2个外显子和 3~9个保守基序,其中,10个为酸性蛋白,6个为碱性蛋白,仅LjCBF1.1蛋白稳定性较好.系统发育树显示,61个LjCBFs分布于Group I,LjCBF1.6位于Group II.组织特异性表达分析显示,LjCBF1.1、LjCBF1.5、LjCBF1.10和 LjCBF1.12在叶、茎、芽和花等 9种组织中均有表达.qRT-PCR检测结果表明,LjCBF1.2和LjCBF1.8受高温、低温、盐、干旱、渗透和LiCl 6种非生物胁迫诱导且显著上调表达.[结论]金银花全基因组中包含 16个CBF/DREB1基因家族成员,其中,LjCBF1.1、LjCBF1.5、LjCBF1.10和LjCBF1.12在金银花不同发育阶段的 9种组织中均有表达,表明其可能对金银花的生长发育具有重要作用;LjCBF1.2和LjCBF1.8显著响应 6种不同非生物胁迫,且在多个组织中表达,推测其参与响应金银花非生物胁迫和生长发育调控过程.
[Objective]The structure,composition and characteristics of CBF/DREB1 gene family members of honeysuckle,and their expression patterns in different tissues and under abiotic stress were investigated to provide a theoretical basis for improving the stress resistance,yield and quality of honeysuckle.[Method]The Blast matching were conducted in the protein database of honeysuckle by using a bioinformatics method and taking the amino acid sequences of 6 CBF/DREB1 family members of Arabidopsis thaliana as templates,and the physicochemical properties and characteristics of LjCBFs were analyzed.The expression patterns of LjCBFs in different tissues were analyzed based on the transcriptome data in the NCBI database.The seedlings of Jinhua 3(a honeysuckle variety)grew under different abiotic stress respectively,and the flower buds were collected to analyze the expression patterns of LjCBFs.[Result]16 CBF/DREB1 gene family members identified from honeysuckle all were hydrophilic proteins,did not contain transmembrane domains and signal peptides,and contained 1-2 exons and 3-9 conserved motifs.Among them,10 were acidic proteins and 6 were basic proteins.The LjCBF1.1 protein had a relatively good stability only.The phylogenetic tree analysis showed that 61 LjCBFs were distributed in Group I,while LjCBF1.6 was located in Group II.The tissue-specific expression analysis showed that LjCBF1.1,LjCBF1.5,LjCBF1.10 and LjCBF1.12 were expressed in 9 tissues including leaves,stems,shoots,flowers and so on.The qRT-PCR detection analysis showed that LjCBF1.2 and LjCBF1.8 were significantly upregulated in response to 6 different abiotic stresses including high temperature,low temperature,salt,drought,osmosis and LiCl.[Conclusion]There are 16 CBF/DREB1 gene family members in the whole-genome of honeysuckle.Among them,LjCBF1.1,LjCBF1.5,LjCBF1.10 and LjCBF1.12 were expressed in 9 tissues at different developmental stages of honeysuckle,which indicates that they may play an important role in growth and development of honeysuckle.LjCBF1.2 and LjCBF1.8 show significant responses to six different abiotic stresses and are expressed in multiple tissues.It is speculated that they are involved in the response to abiotic stresses and growth and development regulation processes of honeysuckle.
