目的:探讨电针"内关"和"足三里"对大脑中动脉栓塞(MCAO)模型大鼠大脑皮层轴突再生中生长相关蛋白43(GAP-43)、神经生长抑制蛋白A(Nogo-A)及其受体(NgR)信号通路的影响.方法:将健康雄性SPF级大鼠随机分为假手术组、模型组和电针组,模型组和电针组采用Zea Longa改良线栓法制备大鼠MCAO模型;假手术组仅暴露颈动脉,不进行动脉闭塞处理.电针组大鼠于造模后24 h进行电针治疗,穴取双侧内关穴、足三里穴,每天电针1次,每次20 min.假手术组与模型组不予任何干预措施.各组分别于治疗第3 d、7 d、14 d后取大鼠大脑组织留作化验.新鲜取材后,以蛋白质印迹(Western Blot)法检测GAP-43、Nogo-A、NgR蛋白的表达,以逆转录聚合酶链式反应(RT-PCR)技术检测Nogo-A mRNA、NgR mRNA的表达.结果:与模型组比较,电针组在治疗后第3 d、第7 d、第14 d的GAP-43表达均升高(P<0.05),而Nogo-A蛋白的表达及Nogo-A mRNA、NgR mRNA的表达均降低(P<0.05),NgR蛋白表达在治疗后第14 d下降(P<0.05).结论:电针"内关"和"足三里"可抑制梗死侧大脑皮层Nogo-A/NgR信号通路相关蛋白及其mRNA表达,上调梗死侧大脑皮层GAP-43的表达,从而减轻神经功能损害.

Objective:To explore the effects of electroacupuncture at"Neiguan"and"Zu sanli"on the growth asso-ciated protein-43(GAP-43)and the Nogo-A/NgR signaling pathway(including neurotrophic inhibitory proteins and their receptors)in axonal regeneration within the infarcted cerebral cortex of rats with middle cerebral artery occlusion(MCAO).Methods:Healthy male SPF rats were randomly divided into a sham operation group,a model group,and an electroacupuncture group.The model group and the electroacupuncture group were used to establish the MCAO model in rats using the Zea Longa modified suture method,while in the sham operation group,only the artery was exposed without occlusion.In the electroacupuncture group,rats received electroacupuncture treatment at bilateral Neiguan and Zu sanli acupoints for 20 minutes once daily,starting 24 hours after model establishment.The sham operation group and the model group received no intervention.Brain tissue was collected from each group for analysis at 3 days,7 days,and 14 days after treatment.After fresh tissue collection,protein expression of GAP-43,Nogo-A,and NgR was detected by Western blot,and the mRNA expression of Nogo-A and NgR were detected by reverse transcription-polymerase chain reaction(RT-PCR).Results:Compared with the model group,the electroacupuncture group showed increased GAP-43 expression at days 3,7,and 14 after treatment(P<0.05),while the protein expression of Nogo-A and the mRNA expression of Nogo-A and NgR were decreased(P<0.05),the protein expression of NgR is dencreased at days 14.Conclusion:Electroacupuncture at"Neiguan"and"Zu sanli"can inhibit the expression of Nogo-A/NgR signaling pathway-related proteins and their mRNA in the infarcted cerebral cortex,and upregulate the expression of GAP-43 in the infarcted cerebral cortex,thereby reducing nerve damage.
