Functions of Slit3 and its role in tumors
轴突导向分子Slit3(Slit guidance ligand 3)作为神经轴突导向分子家族中的一员,不仅引导神经发育,还广泛参与调控血管生成、细胞迁移及器官发育等多个生理过程.近年来研究发现,Slit3在多种肿瘤中表达下调,其下调可能与启动子高甲基化及微RNA(microRNA,miRNA)调控相关,并通过与Robo受体结合,参与调控肿瘤细胞增殖、迁移、侵袭、上皮-间充质转化等过程,进而影响肿瘤的进展与治疗.此外,Slit3可作为多种肿瘤的预后评估、早期诊断及分子分型的潜在生物标志物,也有望成为肿瘤靶向治疗的新靶点,但相关诊疗应用仍需更多临床试验验证.深入理解Slit3的功能及其在多种肿瘤中的作用机制,可为其临床应用提供理论依据,并为靶向Slit3的肿瘤治疗策略提供新的研究方向.
Slit guidance ligand 3(Slit3),a member of the axon guidance molecule family,not only plays a critical role in neural development,but also participates in a variety of physiological processes,including angiogenesis,cell migration,and organ development.Recent studies have demonstrated that Slit3 is downregulated in multiple types of tumors,which may be associated with promoter hypermethylation and microRNA-mediated regulation.By binding to Robo receptors,Slit3 is involved in regulating tumor cell proliferation,migration,invasion,and epithelial-mesenchymal transition,thereby affecting tumor progression and therapeutic response.In addition,Slit3 has the potential to serve as a biomarker for prognosis assessment,early diagnosis,and molecular classification in various cancers,and may also represent a novel target for tumor-targeted therapy.However,its clinical application still requires further validation through large-scale clinical studies.A deeper understanding of the biological functions of Slit3 and its mechanisms in different tumors may provide a theoretical basis for its clinical application and offer new directions for Slit3-targeted therapeutic strategies.
