mt-Pro 5'tiRNA在C2C12细胞增殖中的作用及靶基因预测OA
Effect of mt-Pro 5'tiRNA on the proliferation of C2C12 cells and the prediction of target genes
tiRNAs(tRNA 来源的应激诱导 RNA)在多种疾病的发生发展中起到关键作用,已知线粒体脯氨酸tRNA来源的mt-Pro 5'tiRNA在骨骼肌中表达丰度较高,但其是否在小鼠骨骼肌损伤修复和再生中发挥作用尚不明确.以C2C12 为实验材料,利用 RT-qPCR法检测mt-Pro 5'tiRNA在细胞增殖分化过程中的表达变化,CCK-8 法检测mt-Pro 5'tiRNA对细胞活力的影响,并预测其候选靶基因.结果表明,mt-Pro 5'tiRNA在 C2C12 细胞增殖过程中表达降低,在 C2C12 细胞分化过程中表达升高;过表达mt-Pro 5'tiRNA可降低细胞活力.生物信息学预测共得到 2 273 个候选靶基因,KEGG 通路分析显示,靶基因显著富集于 Hippo 信号通路、Wnt 信号通路及 MAPK 信号通路.研究结果为后续mt-Pro 5'tiRNA在肌肉损伤修复中的作用和机制研究奠定基础.
tiRNAs(stress-inducible RNAs derived from tRNA)play a pivotal role in the pathogenesis and progression of various diseases.It is known that mitochondrial proline tRNA-derived mt-Pro 5'tiRNA exhibits high expression levels in skeletal muscle,yet its potential involvement in the repair and regeneration of skeletal muscle injury in mice remains unclear.C2C12 cells were used as experimental material.The expression changes of mt-Pro 5'tiRNA during cell proliferation and differentiation were detected using RT-qPCR,while the effects of mt-Pro 5'tiRNA on cell viability were assessed by the CCK-8 assay.Additionally,candidate target genes were predicted.The results demonstrated that the expression of mt-Pro 5'tiRNA was reduced during C2C12 cell proliferation but increased during C2C12 cell differentiation.Overexpression of mt-Pro 5'tiRNA decreased cell viability.Bioinformatics analysis identified 2 273 candidate target genes,and KEGG pathway analysis revealed significant enrichment of these genes in the Hippo signaling pathway,Wnt signaling pathway,and MAPK signaling pathway.The findings lay the foundation for further mechanistic studies on the role of mt-Pro 5'tiRNA in muscle injury repair.
