水稻根际促生菌微胶囊菌剂的制备及促生作用OA

Preparation and growth-promoting effects of microcapsules of rice rhizosphere growth promoting bacteria

为推动植物根际促生菌复合菌剂在农业生产中的广泛应用,减少化肥施用,实现农业绿色可持续发展,以解磷菌Pantoea eucalypti R8、解钾菌Enterobacter sichuanensis K25和固氮菌Bacillus zanthoxyli N2 为供试菌株,通过单因素实验、正交实验及响应面法优化复合菌剂发酵条件,采用海藻酸钠-CaCl2制备复合菌剂微胶囊,并通过盆栽实验明确其对水稻(Oryza sativa L.)的促生作用.结果表明,复合菌剂最优发酵培养基为糖蜜20.00 g·L-1,玉米浆粉 20.00 g·L-1,酵母浸膏3.00 g·L-1,K2HPO4 1.50 g·L-1,NaCl 3.00 g·L-1;最优发酵条件为31.0℃,pH 6.95,接种量3.3%,装液量 30.0 mL;优化后发酵液活菌浓度达1.02×1012 cfu·mL-1,较未优化提升 5.05 倍.制备微胶囊最佳配比为 3%SA-3%CaCl2,包埋率达 86.74%,具有良好的缓释性能.盆栽实验中,施用复合菌剂微胶囊的水稻株高、根长、干物质积累及单株产量均显著高于对照组(P<0.05),其中株干物质质量提高 67.86%,单株产量提高22.45%.制备的水稻复合菌剂微胶囊促生作用显著,具有良好的应用潜力,可为微生物肥料制备与应用提供理论和技术支持,对推动绿色农业发展具有积极作用.

To promote the widespread application of plant growth-promoting rhizobacteria composite microbial agents in agricultural production,reduce fertilizer use,and achieve green,sustainable agricultural development,used phosphate-solubilizing Pantoea eucalypti R8,potassium-solubilizing Enterobacter sichuanensis K25,and nitrogen-fixing Bacillus zanthoxyli N2 as research strains.The fermentation conditions for the composite microbial agents were optimized using single-factor,orthogonal,and response surface methodology.Microcapsules were prepared using sodium alginate and CaCl2,and their growth-promoting effect on rice was verified in pot experiments.The results showed that the optimal fermentation medium for the composite microbial agent was molasses 20.00 g·L-1,corn steep powder 20.00 g·L-1,yeast extract 3.00 g·L-1,K2HPO4 1.50 g·L-1,NaCl 3.00 g·L-1.The optimal fermentation conditions were temperature 31.0℃,pH 6.95,inoculation volume 3.3%,and liquid loading volume 30.0 mL.The bacterial concentration reached 1.02×1012 cfu·mL-1 after optimization,which was 5.05 times higher than in the unoptimized state.The optimal preparation ratio for microcapsule was 3%SA-3%CaCl2,with an embedding rate of 86.74%and good sustained-release performance.In the pot experiment,the rice treated with the microcapsule showed significantly higher plant height,root length,dry weight,and single-plant yield than the control group(P<0.05),with 67.86%increase in plant dry weight and 22.45%increase in single-plant yield.The rice composite microbial agent microcapsules prepared exhibited significant growth-promoting effects and great application potential,providing theoretical and technical support for the preparation and application of microbial fertilizers,and plays a positive role in promoting the development of green agriculture.
