为了开发深加工肉制品肉松和肉丸中非洲猪瘟病毒的微滴式数字PCR检测方法,着眼于研究不同提取方法对非洲猪瘟病毒DNA提取效率的影响.通过比较酚氯仿抽提DNA手工法、TaKaRa MiniBEST Viral RNA/DNA Extraction Kit快速提取法、QIAGEN DNeasy○R mericon Food Kit快速提取法和TIANGEN Namp Genomic DNA Kit快速提取法对非洲猪瘟病毒DNA提取效果的影响,发现TaKaRa MiniBEST Viral RNA/DNA Extraction Kit快速提取试剂盒和QIAGEN DNeasy○R mericon Food Kit快速提取试剂盒的效果较为良好.综合提取程序的复杂程度和加热裂解过程的影响,确定TaKaRa MiniBEST Viral RNA/DNA Extraction Kit快速提取法为最适方案.

In order to develop a droplet digital PCR method for detection of African swine fever virus in deep processed meat products such as meat floss and meatballs,the aim was to study the effect of different extraction methods on the extraction efficiency of African swine fever virus DNA.By comparing the effects of different nucleic acid extraction methods on the droplet digital PCR detection re-sults of the virus.The methods evaluated included the phenol-chloroform DNA extraction method,the rapid extraction method using the TaKaRa MiniBEST Viral RNA/DNA Extraction Kit,the rapid extraction method using the QIAGEN DNeasy○R mericon Food Kit,and the rapid extraction method using the TIANGEN Namp Genomic DNA Kit.TaKaRa MiniBEST Viral RNA/DNA Extraction Kit and QIAGEN DNeasy○R mericon Food Kit have a relatively good effect.Based on the complexity of the extraction program and the influ-ence of the heating and cracking process,it was ultimately confirmed that the TaKaRa MiniBEST Viral RNA/DNA Extraction Kit rapid extraction method is the most suitable solution.
