Optimization of Enzymatic Hydrolysis Process for Rhodotorula mucilaginosa and Its Fermentation Product Application in Production of Laying Hens

本试验旨在研究胶红酵母(Rhodotorula mucilaginosa)的酶解工艺优化及其发酵产物在蛋鸡生产中的应用.体外酶解试验采用单因素试验,研究复合酶(酸性甘露聚糖酶和 β-葡聚糖酶)添加量和添加比例、酶解时间、酶解温度和酶解 pH 对胶红酵母突变菌株 CS-15 类胡萝卜素提取率的影响.利用 Box-Behnken 响应面法进一步优化酶解法提取胶红酵母类胡萝卜素的工艺条件.选取 22 周龄、体况相近的海兰褐蛋鸡 180 只,随机分为 3 组,每组 6 个重复,每个重复10 只鸡.A 组(对照组)饲喂基础饲粮,B 组和 C 组分别在基础饲粮中添加 1%和 2%胶红酵母发酵产物.动物试验的预试期为 3 d,正试期为 28 d.结果表明:1)复合酶的最佳添加比例为酸性甘露聚糖酶∶β-葡聚糖酶=1∶2,最佳添加量为 100 mg/L.在此添加比例和添加量下,类胡萝卜素提取率达 63.14%.最佳工艺条件为:酶解时间 5 h、酶解 pH 4.5、酶解温度 50℃.在此条件下,类胡萝卜素提取率预测值为 75.51%,实际提取率为(74.50±0.18)%.2)B 组和 C 组在各时间点的蛋黄 Roche 色度等级和类胡萝卜素含量均显著高于对照组(P<0.05),且 C 组显著高于 B 组(P<0.05).各组之间产蛋率、蛋壳强度、蛋壳厚度等无显著差异(P>0.05).由此可见,本试验对复合酶酶解提取胶红酵母类胡萝卜素的工艺条件进行了优化,并通过动物试验证实胶红酵母发酵产物可安全替代合成添加剂,有效改善鸡蛋蛋黄色泽和营养价值.

This experiment was conducted to study the optimization of enzymatic hydrolysis process for Rhodotorula mucilaginosa and its fermentation product application in production of laying hens.The in vitro en-zymatic hydrolysis experiment used a single-factor experiment,to study the effects of compound enzyme(acid-ic mannanase and β-glucanase)addition amount and addition ratio,enzymatic hydrolysis time,enzymatic hy-drolysis temperature and enzymatic hydrolysis pH on carotenoid extraction rate of Rhodotorula mucilaginosa mutant strain CS-15.Further optimization of process conditions for carotenoids from Rhodotorula mucilaginosa by enzymatic method using Box-Behnken response surface methodology.A total of 180 Hy-Line brown laying hens aged at 22 weeks with similar body condition were randomly assigned to 3 groups with 6 replicates per group and 10 hens per replicate.The laying hens in group A(control group)were fed a basal diet,and others in group B and group C were fed basal diets supplemented with 1%and 2%Rhodotorula mucilaginosa fermen-tation product,respectively.The pre-experimental period lasted for 3 days,and the experimental period lasted for 28 days.The results showed as follows:1)the optimal addition ratio of compound enzyme was acidic mannanase∶β-glucanase=1∶2,and the optimal addition amount was 100 mg/L.Under the addition ratio and addition amount,the carotenoid extraction rate reached 63.14%.The optimal process conditions were hydroly-sis time of 5 h,hydrolysis pH of 4.5 and hydrolysis temperature of 50℃.Under these conditions,the predic-ted carotenoid extraction rate was 75.51%,and the actual extraction rate was(74.50±0.18)%.2)The yolk Roche color level and carotenoid content at various time points of group B and group C were significantly high-er than those of the control group(P<0.05),and group C was significantly higher than group B(P<0.05).There were no significant differences in egg production rate,eggshell strength and eggshell thickness et al a-mong all groups(P>0.05).In conclusion,the experiment optimizes the conditions for extracting carotenoids from Rhodotorula mucilaginosa by enzymatic hydrolysis using compound enzyme,and through animal experi-ments,it has been confirmed that the Rhodotorula mucilaginosa fermentation product can safely replace syn-thetic additives,effectively improve the yolk color and nutritional value of eggs.[Chinese Journal of Animal Nutrition,2026,38(4):2651-2662]
