岩藻多糖对过敏性腹泻断奶仔猪生长性能、腹泻、肠道通透性和抗氧化能力的影响OA
Effects of Fucoidan on Growth Performance,Diarrhea,Intestinal Permeability and Antioxidant Capacity of Weaned Piglets with Allergic Diarrhea
本试验通过在饲粮中添加大豆抗原蛋白建立断奶仔猪过敏性腹泻模型并饲喂岩藻多糖,探讨岩藻多糖对断奶仔猪过敏性腹泻的缓解作用.试验选取 40 头断奶仔猪,随机分为 5组,每组 8 头仔猪(n=8),分别饲喂基础饲粮(CT 组)、基础饲粮+2%大豆抗原蛋白(SAP 组)、基础饲粮+2%大豆抗原蛋白+100 mg/kg 岩藻多糖(SAP+LF 组)、基础饲粮+2%大豆抗原蛋白+200 mg/kg 岩藻多糖(SAP+MF 组)、基础饲粮+2%大豆抗原蛋白+300 mg/kg 岩藻多糖(SAP+HF 组),进行为期 6d 的饲养试验.结果表明:1)与 CT 组相比,饲粮中添加大豆抗原蛋白显著降低了整个试验期内断奶仔猪的平均粪便评分(P<0.05),并显著增加了严重腹泻天数(P<0.05);与 SAP 组相比,饲粮中添加 200 mg/kg 岩藻多糖显著提高了平均粪便评分(P<0.05),添加 100、200 和 300 mg/kg 岩藻多糖均显著降低了腹泻率(P<0.05).2)与 CT 组相比,饲粮中添加大豆抗原蛋白显著增加了血清中免疫球蛋白 E(IgE)和 D-乳酸(D-LA)含量以及空肠中糜蛋白酶活性(P<0.05);与 SAP 组相比,饲粮中添加 100、200 和 300 mg/kg 均显著降低了血清中IgE 含量(P<0.05).3)与 CT 组相比,饲粮中添加大豆抗原蛋白显著降低了空肠和回肠的绒毛高度和绒毛高度与隐窝深度比值(P<0.05),并显著增加了空肠的隐窝深度(P<0.05);与 SAP组相比,饲粮中添加 300 mg/kg 岩藻多糖显著增加了空肠和回肠的绒毛高度(P<0.05),饲粮中添加 200 mg/kg 岩藻多糖显著降低了空肠的隐窝深度(P<0.05),饲粮中添加 200 和 300 mg/kg岩藻多糖还显著提高了空肠的绒毛高度/隐窝深度比值(P<0.05).4)与 SAP 组相比,饲粮中添加 300 mg/kg 岩藻多糖显著上调了空肠中闭锁小带蛋白-1(ZO-1)、密封蛋白-1(Claudin-1)和闭合蛋白(Occludin)的 mRNA 相对表达量(P<0.05).5)与 CT 组相比,饲粮中添加大豆抗原蛋白显著降低了血清中过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)活性(P<0.05),并显著降低了空肠中 CAT 活性(P<0.05);与 SAP 组相比,饲粮中添加 200 和 300 mg/kg 岩藻多糖显著提高了血清中 CAT 和 GSH-Px 活性(P<0.05),饲粮中添加 200 mg/kg 岩藻多糖还显著提高了空肠中 CAT 活性(P<0.05).此外,大豆抗原蛋白显著下调了空肠中锰超氧化物歧化酶(Mn-SOD)的表达(P<0.05),而 200 mg/kg 岩藻多糖显著上调了空肠中 CAT、铜锌超氧化物歧化酶(CuZn-SOD)、Mn-SOD、谷胱甘肽过氧化物酶 1(GPx1)和谷胱甘肽过氧化物酶 4(GPx4)的表达(P<0.05).综合而言,饲粮中添加岩藻多糖能够改善大豆抗原蛋白导致的空肠和回肠黏膜损伤,抑制肠道通透性的增加,上调紧密连接蛋白和抗氧化酶基因表达,降低过敏反应诱导的抗氧化能力下降,缓解断奶仔猪的过敏性腹泻.
In this experiment,a model of allergic diarrhea in weaned piglets was established by soy antigen protein(SAP)and feeding fucoidan to investigate the alleviating effect of fucoidan on allergic diarrhea in weaned piglets.A total of 40 weaned piglets were randomly assigned to 5 groups with 8 piglets per group(n=8).The piglets in the 5 groups were fed different diets:basal diet(CT group),basal diet+2%SAP(SAP group),basal diet+2%SAP+100 mg/kg fucoidan(SAP+LF group),basal diet+2%SAP+200 mg/kg fu-coidan(SAP+MF group),basal diet+2%SAP+300 mg/kg fucoidan(SAP+HF group).The feeding trial was lasted for 6 days.The results showed as follows:1)compared with the CT group,dietary supplementation with SAP significantly reduced the average fecal score throughout the experimental period(P<0.05),and sig-nificantly increased the severe diarrhea days of weaned piglets(P<0.05).Compared with the SAP group,diet-ary supplementation with 200 mg/kg fucoidan significantly increased the average fecal score(P<0.05),and supplementation with 100,200 and 300 mg/kg fucoidan significantly reduced the diarrhea rate(P<0.05).2)Compared with the CT group,dietary supplementation with SAP significantly increased the contents of serum immunoglobulin E(IgE)and D-lactic acid(D-LA)as well as jejunal chymotrypsin activity(P<0.05).Com-pared with the SAP group,dietary supplementation with 100,200 and 300 mg/kg fucoidan significantly re-duced the serum IgE content(P<0.05).3)Compared with the CT group,dietary supplementation with SAP significantly decreased the villus height and the villus height to crypt depth ratio in the jejunum and ileum(P<0.05),and significantly increased the crypt depth in the jejunum(P<0.05).Compared with the SAP group,dietary supplementation with 300 mg/kg fucoidan significantly increased the villus height in the jejunum and il-eum(P<0.05),dietary supplementation with 200 mg/kg fucoidan significantly decreased the crypt depth in the jejunum(P<0.05),and dietary supplementation with 200 and 300 mg/kg fucoidan also significantly in-creased the villus height to crypt depth ratio in the jejunum(P<0.05).4)Compared with the SAP group,diet-ary supplementation with 300 mg/kg fucoidan significantly upregulated the mRNA relative expression levels of zonula occludens-1(ZO-1),Claudin-1 and Occludin in the jejunum(P<0.05).5)Compared with the CT group,dietary supplementation with SAP significantly reduced the activities of catalase(CAT)and glutathione peroxidase(GSH-Px)in serum(P<0.05),and also significantly reduced CAT activity in the jejunum(P<0.05).Compared with the SAP group,dietary supplementation with 200 and 300 mg/kg fucoidan significantly increased serum CAT and GSH-Px activities(P<0.05),and dietary supplementation with 200 mg/kg fucoidan also significantly increased CAT activity in the jejunum(P<0.05).Furthermore,soybean antigen protein sig-nificantly downregulated the expression of manganese superoxide dismutase(Mn-SOD)in the jejunum(P<0.05),whereas 200 mg/kg fucoidan significantly upregulated the expression of CAT,copper-zinc superoxide dismutase(CuZn-SOD),Mn-SOD,glutathione peroxidase 1(GPx1)and glutathione peroxidase 4(GPx4)in the jejunum(P<0.05).In conclusion,dietary supplementation with fucoidan can alleviate jejunal and ileal mu-cosal damage induced by SAP,inhibit the increase in intestinal permeability,upregulate the expression of tight junction protein and antioxidant enzyme genes,attenuate the decline in antioxidant capacity introduced by aller-gic reactions,and mitigate allergic diarrhea in weaned piglets.[Chinese Journal of Animal Nutrition,2026,38(4):2534-2546]
