为解析江淮地区大豆种质资源蛋白质与油脂含量的遗传基础,助力优质大豆分子育种,本研究以 320 份江淮大豆种质为材料,于2017-2021 年不同月份开展随机区组试验,测定籽粒蛋白质含量、油脂含量表型数据,利用 61 174 个高质量 SNP 位点,通过 CMLM 模型进行全基因组关联分析(Genome-Wide Association Study,GWAS),结合单倍型分析挖掘优势位点与候选基因.结果显示:两个性状均呈正态分布,符合数量性状遗传特征;蛋白质含量广义遗传率为55.6%,油脂含量为69.3%,后者受环境干扰更小.共筛选出6 个与蛋白质含量显著相关的 SNP 位点(-log10 P≥4.78),其中 Gm20_30617410 为多环境重复位点;检测到 17 个油脂含量显著相关 SNP 位点,在 E3、E4、E6 和 E7 这4 个 环境中重复定位到 12 个位点,集中于 Gm05 染色体.候选基因挖掘显示,Glyma20g21190(核糖体蛋白)、Glyma20g21540(氨基转移酶)等可能调控蛋白质合成,Glyma05g33190(氧化还原酶)、Glyma05g33360(短链脱氢酶)等参与油脂代谢.研究结果可为大豆品质性状的分子标记辅助育种提供了重要理论依据.

To clarify the genetic basis of protein and oil contents in soybean germplasm resources from the Jianghuai region and facilitate molecular breeding of high-quality soybeans,this study used 320 soybean germplasm accessions from the Jianghuai region as materials.Field experiments were conducted under a randomized complete block design across different months from 2017 to 2021 to determine the phenotypic data of seed protein content and oil content.A total of 61 174 high-quality SNP markers were used for Genome-wide Association Study(GWAS)via the CMLM model,and haplotype analysis was combined to identify favorable loci and candidate genes.The results showed that both traits exhibited normal distribution,consistent with the genetic characteristics of quantitative traits.The broad-sense heritability of protein content was 55.6%,while that of oil content was 69.3%,indicating that the latter was less affected by environmental factors.The result identified 6 SNP markers significantly associated with protein content(-log10P≥4.78),among which Gm20_30617410 was detected repeatedly across multiple environments.17 significant SNP loci were detected for oil content,12 loci were repeatedly mapped in four environments(E3,E4,E6,and E7),and these loci were concentrated on the Gm05 chromosome.Candidate gene mining showed that Glyma20g21190(ribosomal protein)and Glyma20g21540(aminotransferase)might regulate protein synthesis,while Glyma05g33190(oxidoreductase)and Glyma05g33360(short-chain dehydrogenase)were involved in oil metabolism.The significant SNP markers and candidate genes identified in this study provide important theoretical basis and technical targets for marker-assisted breeding of soybean quality traits.
